Objective: By observing the traditional Tibetan features and enzyme activities of theorganization "Pazufloxacin pill" chronic alcoholic liver disease (alcoholic liverdisease, ALD) in rat liver, and to view the impact of Pazufloxacin Pill on alcoholicliver injury in rats with the expression of PPAR-α1, AMPK-α1,and to explore theprotective effect for Pazufloxacin pill on rats with chronic alcoholic liver disease andalso to research its possible mechanism.Methods: The78inbred male SD rats fed for a week, were totally randomly dividedinto normal control group, model group, DDB Group, Pazufloxacin pills low,medium and high dose group of6group, model control group has18rats, Theremaining12rats in each other group. Model group, DDB Group, Pazufloxacin pilllow, medium and high dose groups were given56%of the Red Star Erguotou wine10mL·kg-1gavage; given normal saline10mL·kg-1day morning at a time, ig12weeks, manufacturing ethanol-induced liver injury model. After the modeling,Pazufloxacin pills low, medium and high dose groups were given10mL·kg-1pillsuspension Pazufloxacin gavage doses were0.05,0.10,0.20g·kg-1; DDB GroupDDB dripping pills give10mL·kg-1suspension orally at a dose of0.003g·kg-1;normal control group, model control group were given the same volume of saline ineach group fed a daily morning times gavage three weeks.24h ban water fasting,anesthesia, anatomy, sampling. Serum alanine aminotransferase (ALT), aspartateaminotransferase (AST), triglyceride (TG), superoxide dismutase (SOD), reducedglutathione (GSH), glutathione over peroxidase (GSH-Px), malondialdehyde (ADM)level of activity. And detecting liver PPAR-α1mRNA, expression levels ofAMPK-α1mRNA.Results: Contrast with the normal group, model group, serum liver wet weight andliver index and ALT, AST, TG, MDA were markedly increased (P <0.05), SOD,GSH, GSH-Px were markedly lower (P <0.05), liver tissue model group decreasedsignificantly PPAR-α1mRNA, AMPK-α1mRNA (P <0.01); contrast with themodel group, the Pazufloxacin pills each dose group and the DDB group liver wetweight and liver index and ALT, AST, TG were markedly lower (P <0.05), markedlyreduced MDA (P <0.01), SOD, GSH, GSH-Px were markedly increased (P <0.05); contrast with the model group, the Pazufloxacin pills each dose group and DDBgroup PPAR-α1mRNA, AMPK-α1mRNA were significantly increased (P <0.05).Conclusion: Pazufloxacin pill can effectively protect ethanol on rat liver damage,improve liver function, reduce triglyceride levels, and inhibiting lipid peroxidationinduced by ROS. suggesting the drug on ethanol-induced liver disease have aprotective effect.
