Objective:To study the protective effect and mechanism of Gardenia jasminoides Ellis aqueous extract?SZZJYA?on immunological hepatic injury and alcoholic hepatic injury in mice.Methods:?1?Effect of SZZJY on anti-immunological hepatic injury:Kunming mice were randomly divided into six groups:NC group,model group(20mg·kg-1),bifendate group(150mg·kg-1,BPDC),high?equal to crude drug40g·kg-1?,medium?equal to crude drug 20g·kg-1?and low?equal to crude drug10g·kg-1?dose group of SZZJYA.NC group and liver-injured group were intragastrically administered with distilled water,the other four groups were given the corresponding drug once a day for 12 days by i.g.60 mins after the last administration,NC group were injected with physiological saline via tail vein,and the other groups were injected with concanavalin A via tail vein to establish immunological hepatic injury model.Then all the mice were killed after 8hrs.Serum and liver tissues were collected,and the coefficient of the liver,spleen,thymus were determined.The activity of ALT,AST,ALP,TBIL,ALB,GLB and A/G in serum,the concentration of MDA,GSH in liver tissues and the activity of SOD and GSH-PX were investigated.HE stain was used to determine the degree of hepatic injury and immunohistochemical method was used to determine the expressions of Bax and Bcl-2 in liver tissues.Enzyme-linked immunosorbent assay was used to test the expressions of TNF-?,IFN-?and IL-4 in serum.?2?Effect of SZZJYA on anti-alcoholic hepatic injury:Kunming mice were randomly divided into six groups:NC i.g,and the other groups were given the 52%vol star erguotou liquor by i.g to establish a model of alcoholic hepatic injury,once a day for seven days.After the last administration,all the mice were killed after 16hrs.Serum and liver tissues were collected,and the coefficient of the liver was determined.The activities of ALT,AST,ALP,TBIL,TP,ALB,GLB and A/G in serum,the concentration of MDA and GSH in hepatic homogenate,and the activities of SOD and GSH-PX were investigated.HE stain was used to determine the degree of hepatic injury and immunohistochemical method was used to determine the expression of NF-?B and TNF-?in liver tissue.Results:?1?Effect of SZZJYA on anti-immunological hepatic injury:Compared with model group,different doses of SZZJYA and positive control group could improve weight and the coefficient of the spleen,and reduce abnormal elevation of TBIL which induced by ConA in mouse serum,and significantly increased the activity of SOD in hepatic homogenate?P<0.01 or P<0.05?.SZZJYAH and SZZJYAM could decrease the serum transaminase and ALP activity and the MDA concentration in hepatic homogenate,and increase the TP and ALB concentration in mice serum?P<0.01 or P<0.05?SZZJYAH and SZZJYAM could also increase the GSH,GSH-PX activities in hepatic homogenate?P<0.01 or P<0.05?.At the same time,SZZJYAM increased the GLB concentration?P<0.05?in mouse serum.Compared with the model group,positive control group could obviously raise the A/G?P<0.05?but there was no significant difference in different doses of SZZJYA with A/G.Compared with model group,different doses of SZZJYA and positive control group could be significantly lessened the degree of hepatic injury.The expression of Bax was obviously reduced and the expression of Bcl-2 was increased in hepatic homogenate?P<0.05 or P<0.01?after SZZJYA and positive drug treatment.Compared with model group,different doses of SZZJYA and positive control group could markedly reduce the expressions of TNF-?.SZZJYAH,SZZJYAM could markedly reduce the expressions of TNF-?and IL-4.?2?Effect of anti-alcoholic hepatic injury:compared with model group,SZZJYAH and SZZJYAM and positive control groups could markedly improve weight and reduce the coefficient of the liver?P<0.05?,but the abnormal elevation could induce by alcohol of the activity of AST,ALT and ALP,and the content of TBIL in the mice serum?P<0.01 or P<0.05?could be obviously reduced,and significantly increase the content of ALB and TP?P<0.01?at different doses of SZZJYA and positive control groups.Among them,SZZJYAM could increase the ALP activity?P<0.01orP<0.05?.In addition,SZZJYAH and SZZJYAM and positive control groups reduced the concentration of MDA in hepatic homogenate?P<0.01 or P<0.05?and different doses of SZZJYA significantly increased the activity of SOD and GSH?GSH-PX?P<0.01 or P<0.05?.Compared with model group,the degree of hepatic injury of different doses of SZZJYA and positive control groups could significantly lessen the degree of hepatic;Among them,SZZJYAH and SZZJYAM could markedly decrease the expression of NF-?B and TNF-??P<0.01 or P<0.05?.Conclusion:SZZJYA show protective effects on immunological and alcoholic hepatic injury in mice.The mechanism may be related to the inhibition of cytokine expression,attenuating oxide free radical,lipid peroxidation and apoptosis.
