Expression And Significance Of Hsa_circ₀000432 And Hsa_circ₀000690 In Breast Cancer Serum

Objective:We collected serum from breast cancer patients and healthy people to detected the expression of hsa_circ₀000432 and hsa_circ₀000690,then explored the relationship between them and the clinical pathological factors of breast cancer patients.Finally,analyzed the clinical value of these two circular RNAs in the early diagnosis and treatment of breast cancer.Methods :1.The serum samples were collected from 50 breast cancer patients who were admitted to the First Affiliated Hospital of University of south china?their postoperative pathological results were definitely diagnosed with breast cancer,and without radiotherapy or chemotherapy?.Then we collected 50 serum samples of female healthy people whose age matched with breast cancer patients?Their medical report was free of malignant tumors,breast lumps and chronic diseases of important systems?;2.The Quantitative Real-time PCR mehhod was used to detect the expression of hsa_circ₀000432 and hsa_circ₀000690 in the serum of 50 breast cancer patients and 50 healthy people.Then,compared the difference in expression of these two circular RNAs in breast cancer and healthy people;3.To analyze the relationship between the expression levels of serum hsa_circ₀000432 and hsa_circ₀000690 in breast cancer patients and the clinical pathological factors of breast cancer;4.Receiver operating characteristic?ROC?curve used to evaluate the diagnostic value of hsa_circ₀000432 and hsa_circ₀000690 for breast cancer.Results:1.The expression levels of hsa_circ₀000432 and hsa_circ₀000690 in the serum of breast cancer patients were lower than those of healthy people?P<0.05?.2.The expression level of hsa_circ₀000432 was negatively correlated with the age and TNM staging of breast cancer patients?P <0.05?,but not related to the patient's tumor size,tumor differentiation degree,lymph node,ER,PR,HER2,etc.?P> 0.05?.The expression level of hsa_circ₀000690 was correlated with the tumor differentiation degree and TNM staging of breast cancer patients?P<0.05?,and it was positively correlated with the tumor differentiation degree and negatively correlated with TNM stage,but not related to the patient's age,lymph node,tumor size,ER,PR and HER2?P> 0.05?.3.The ROC curve results showed,the AUC of hsa_circ₀000432 was 0.848?95%CI: 0.765 0.931,P<0.05?,and the AUC of hsa_circ₀000690 was 0.659?95% CI: 0.553 0.766,P<0.05?,the AUC of the combined was 0.750?95% CI: 0.682-0.817,P <0.05?.Conclusion: 1.Hsa_circ₀000432 and hsa_circ₀000690 were low expression in the serum of breast cancer patients.2.The expression level of hsa_circ₀000432 was correlated to the age and TNM staging of breast cancer.3.The expression level of hsa_circ₀000690 was related to the tumor differentiation degree and TNM staging of breast cancer.
