abjective: 1.To investigate the expression of c AMP response element binding protein(CREB)with the cancerous tissue and serum in breast cancer and fibroadenoma,and analyze it's relationship with clinical factors,and then further explore its clinical significance.Methods: 1.There were 30 patients respectively in the two group,each of which was the breast cancer patients and the patients who suffer from breast fibroadenoma,and was randomly selected from May to November 2017 in our hospital.(Patients with breast cancer entered the experimental group,who were required to meet the following requirements: complete case information,no treatment before operation,confirmed breast cancer by intraoperative rapid freezing pathology and routine pathological examination in our hospital;Patients with fibroadenoma of the breast entered the control group,all of the whom needed have completely case data and underwent surgery,and finally diagnosed with the breast fibroadenoma via operation and postoperative pathology in our hospital).The next steep was collecting these lesion tissue samples in different groups,and then detecting the content of CREB by using Western Blot technique.Finally we used the Statistical software(SPSS21.0)to analyze the two groups' data by variance analysis and compare the expression of CREB in breast malignant lesion tissues with the one of benign lesion,and meanwhilewe should analyze the clinical significance of the expression of CREB in breast cancer.2.At the same time,we needed take blood samples with patients mentioned above by using the red head blood vessel without anticoagulant,then made them centrifugation after settling.The next steep was cryopreservation and finally used the enzyme linked immunosorbent assay to detect the content of CREB about the two groups of data.When analyzing the data of two groups by using analysis of variance once more,we needed compare and analyze the expression of serum's CREB in benign and malignant breast diseases,and further analyzed the clinical significance of the expression of CREB in serum of breast cancer cases.3.The Pearson product moment correlation was used to analyze the correlation about the expression of CREB in cancerous tissue and serum of the breast cancer patients.4.According to the age of patient,tumor size(T),lymph node metastasis(N),phase,histological grade,and the experession of the other objects of observation,including ER?PR,Her-2,Ki-67,We respectively divided the breast malignant lesion tissues into two groups.The next step is using the t test of independent samples to compared the expression of CREB with all factors and analyzed their clinical significance.(No distant metastasis was found in the clinical data of all patients with breast cancer in this experime-nt,so the "distant metastasis M" was not compared).The same methods and principles were applied to the serum of the patients who suffered from breast cancer.And then we needed analyze these data.Finally if the statistical analysis showed that P was less than 0.05,we can think that thedifference is statistical significant.Results: 1.We found that it has difference about the expression of CREB between breast cancer tissues(n=30,0.431±0.132)and the benign breast disease tissues(n=30,0.136±0.089),and we considered that the difference has a notable statistical significant(P=0.000).2.There was a signifycantly higher level about the expression of CREB in the serum of breast cancer patients(1.481±0.181)than it's in serum of patients with benign breast disease(0.461±0.100),and the difference also has a notable statistical significance(P=0.000).3.We can get a result by the Pearson product moment correlation: the correlation coefficient of the expression of CREB in the cancer tissues and serum of the breast cancer patients was 0.531,indicating that there was a moderate positive correlation,and the P<0.001,indicating that the correlation coefficient was statistically significant.4.According to these objects of observation in the breast cancer tissue in this experiment,including the age of patient,tumor size(T),lymph node metastasis(N),histological grade,ER?PR,Her-2,Ki-67,the result showed that there was no statistical difference when compared the expression of CREB between with the every two groups(all of these P-values were greater than 0.05).5.As same as above,according to these objects of observation in the breast cancer serum in this experiment,including the age of patient,tumor size(T),lymph node metastasis(N),histological grade,ER?PR,Her-2,Ki-67,the result showed that there was no statistical difference when compared the expression of CREB between with the every two groups(all of these P-values were also greater than 0.05).Conclusion:CREB is expressed in the lesion tissues and serum both of benign and malignant breast lesions,and its expression in lesions tissue of breast cancer and serum is significantly higher than that in benign breast tissue.The expression of CREB in breast cancer serum is also higher than that in fibroadenoma of the breast.And there is a significant positive correlation about the expression of CREB between the serum and cancerous in the breast cancer patients,which maybe have a certain significance on screening and diagnosis for different breast diseases.Moreover,in breast cancer patients,the expression of CREB in cancerous tissue and serum may be not related to the age?tumor size?lymph node metastasis?histological grade?ER?PR?Her-2,or Ki-67 level.
