OBJECTIVETo discuss the expression of the tumor makers B7-H4in serum and tissues of patients with breast cancer and analyze the correlation between B7-H4and relative clinical and pathological factors in order to find a new treatment target of breast cancer.METHORDS1.Elisa clip keep method was used to detect soluble B7-H4(Soluble B7-H4, sB7-H4. First the expression levels of Sb7-h4in serum of20healthy volunteers,29patients with benign breast lesions and53patients with breast cancer were detected. Then the differences between the three groups were analysed in order to study the correlation between the expression of sB7-H4in breast cancer group and clinicopathological parameters.2.53paraffin wax specimens after surgical resection of breast cancer group in Elisa clip keep method were collected, and immuno histochemical method SP was used to detect the expression of B7-H4in the53cases. Then according to these data, the correlation between the expression of B7-H4in patients with breast cancer and the clinical parameters was analysed, which further verified the expression level and the clinical significance of B7-H4in patients with breast cancer.RESULTS1.Elisa test results:1)The expression level of sB7-H4in serum of breast cancer group [(52.76±5.75) ug/1] was significantly higher than healthy volunteer group [(9.15±2.19) ug/1] and benign breast lesion group [(34.28±4.56) ug/1](P<0.01). The expression level of sB7-H4was significantly different (P<0.01) among the three groups, and it was progressively elevated with healthy volunteer group, benign lesion group and breast cancer group.2)The expression level of sB7-H4protein in serum of breast cancer group was correlated with histopathological grade and the expression status of Human Epidermalgrowth Factor Receptortype2. The expression levels of sB7-H4in Grade Ⅱ [(54.68±3.43) ug/1] and Grade Ⅲ[(54.96±2.86) ug/1] were both higher than Grade Ⅰ[(43.94±6.19) ug/1], which was significantly different (P<0.01). However, There was no significant difference (P>0.05) between Grade Ⅱ and Grade Ⅲ. Also there was no correlation between the expression level of sB7-H4and patients’age, the tumor size, lymph node metastasis, clinical stage, estrogen receptors status and progestin receptor status (P>0.05)2.Immunohistochemical test results:B7-H4protein was positive expression in almost all the53patients with breast cancer. The expression of B7-H4protein was related to histopathological grade and expression status of HER2(P<0.05), while there is no correlation between it and patients’age, tumor size, lymph node metastasis, clinical stage and expression status of ER and PR (P>0.05)CONCLUSIONS1. B7-H4could help to find and diagnose breast cancer early, and it is expected to be a new serum tumor marker of breast cancer.2. B7-H4could be used to monitor the recurrence of breast cancer or predict the prognosis of patients. Its high expression which would become a new target of antibody targeted therapy is related to the recurrence, the diffusion, the metastasis and the mortality of breast cancer.3. ELISA test and immunohistochemical detection had the same statistical results, which showed the expression of B7-H4in the serum of breast cancer patients did not significantly differ from that in breast cancer tissues, and its expression in the serum of breast cancer patients could reflect its expression in the breast cancer tissue.
