Objective:To evaluate the clinical effect of Chaihu plus keel Oyster Decoction in the treatment of depression and its effect on serum inflammatory factors,and to explore the related factors and mechanism of its antidepressant effect.Methods:1.Clinical study:83 patients with depression of stagnation of liver Qi were selected as the research object.They were randomly divided into the control group(41 patients taking escitalopram oxalate)and the treatment group(42 patients taking Chaihu plus keel oyster Decoction on the basis of the control group),The treatment period of the two groups was 8 weeks.The Hamilton Depression Scale(HAMD)and inflammatory factors were used as evaluation indexes to evaluate the clinical efficacy of Chaihu plus keel oyster Decoction in the treatment of depression and its influence on infl-ammatory factors,and to analyze the influencing factors of HAMD score decrease during the treatment of 3 days.2.Experimental study:SPF male C57BL/6J mice were used as the blank group,and cums depression model mice were prepared at the same time.According to the difference of administration,they were randomly divided into model group,traditional Chinese medicine group and Western medicine group.After 21 days,the depression behavior indexes of mice were sugar water preference experiment,open field experiment,forced swimming experiment,opening experiment,tail suspension experiment and new food inhibition experiment,and the levels of IL-1 ?,IL-6 and TNF-? in serum were measured for statistical analysis.Results:1.Clinical study:The HAMD scores of the treatment group and the control group were significantly lower than before treatment(P<0.01,P<0.05),and the trend lasted until the 8th week,and the level of IL-1,IL-6 and TNF-? in the treatment group and the control group were significantly lower than before treatment,and the treatment group was better than the control group(P<0.05).Further analysis of the relevant factors based on the decrease of HAMD score at the third day of treatment showed that the scores of women,patients with short course of disease and short years of education decreased more significantly(P<0.05).2.Experimental study2.1 At the end of 21 days,it was found that the immobility time of forced swimming in the Chinese medicine group and Western medicine group was significantly shorter than that in the model group(P<0.05,P<0.01),and the immobility time of tail suspension in the Chinese medicine group and Western medicine group was significantly shorter(P<0.05),and there was no significant difference between the two groups in forced swimming and tail suspension experiment(P>0.05).The central exercise time and the total distance of the opening activities in both Chinese medicine group and Western medicine group were significantly prolonged(P<0.01).There was no significant difference between the two groups(P>0.05).2.2 After 21 days of treatment,the content of IL-1?,IL-6 and TNF-? in the traditional Chinese medicine group and the western medicine group were significantly lower than that in the model group(P<0.0001),and the level of IL-6 in the traditional Chinese medicine group was significantly lower than that in the western medicine group(P<0.05).Conclusion:1.Chaihu plus keel Oyster Decoction has a rapid effect in the treatment of depression with liver depression and qi stagnation,especially for women,those with a short course of disease and low education,and can significantly improve the clinical efficacy.Its mechanism may be related to the reduction of inflammatory factors IL-1,IL-6 and TNF-?.2.Chaihu plus keel Oyster Decoction can significantly improve the behavioral indexes of depression mice model,and its mechanism may be related to the decrease of the levels of IL-1?,IL-6 and TNF-?.
