| Part I:Analysis of the regularity of Youguiyin in treating GA-ONFH based on data analysisObjective:To use Excel software and SPSS software analysis treatment of Glucocorticoid-Associated Osteonecrosis of the Femoral Head necrosis of traditional Chinese medicine formula medication regularly.Methods:Traditional Chinese medicine prescriptions for GA-ONFH in PubMed,EMbase,the Cochrane Library,CBM,CNKI,WanFang and Wip over the past 20 years were collected.The syndrome types,syndrome elements,prescriptions,medication frequency and nature and taste of GA-ONFH were analyzed by using Excel software to explore the core components of the drugs.Association rule analysis is conducted with the Apriori template provided by SPSS Clementine 12.0 software.SPSS software was used for systematic cluster analysis and factor analysis of the nature and taste of drugs.Results:a total of 16 hormone type femoral head necrosis syndrome types were collected from 53 articles,among which kidney Yang deficiency,kidney deficiency and blood stasis,Qi stagnation and blood stasis were the main syndrome types.There are 8 syndrome elements,with Kidney Deficiency,Blood Stasis and Phlegm Turbidity as the main syndrome elements.Prescription more than 30 first,right to drink,peach Siwu Decoction,Bushenhuoxue Decoction;The results of rule analysis showed that there were 9 association rules of drug pairs and 60 association rules of 3 traditional Chinese medicine groups.Nine drug groups were obtained by cluster analysis,and the fitness test of factor analysis showed that it was suitable for factor analysis,and a total of 7 common factors were extracted.Association rules reflect the characteristics of the treatment of Tonifying Kidney and Activating Blood.In the treatment of GA-ONFH,the combination was based on Yougui decoction,accompanied by activating blood and removing blood stasis.Cluster analysis and factor analysis showed the characteristics of tonifying kidney and promoting Yang,activating blood circulation and resolving blood stasis.In addition,factor analysis could reflect the characteristics of wenjing sanhan and clearing damp resolving phlegm.Conclusion:the main drugs used in traditional Chinese medicine treatment of glucocorticosteroid-related femoral head necrosis are tonifying the kidney and promoting Yang and activating blood circulation,and most of the drugs used in the treatment of liver,kidney,spleen and stomach meridians are sweet.This result will provide theoretical guidance for the clinical treatment of GA-ONFH in traditional Chinese medicine.Part Two:Bioinformatics Analysis of the Mechanism of Youguiyin in Treating GA-ONFH of"Deficiency of Kidney Yang"Purpose:Based on the bioinformatics method,the possible mechanism of Youguiyin in the treatment of GA-ONFH with kidney-yang deficiency type was analyzed.Methods:1.Mass spectrometry detection of Youguiyinchengfang and its single drugs,screening of active ingredients,and prediction of possible targets;2.Retrieve the target sites related to femoral head necrosis,and construct a network diagram of"active ingredients-target sites" for Youguiyin;3.Construct a network diagram of the interaction between Youguiyin and femoral head necrosis and screen out key targets;4.Perform signal pathway enrichment analysis on key targets to find the main enrichmentpathways of key targets to predict the possible mechanism of Youguiyin treatment of GA-ONFH.Results and conclusions:Youguiyin may mainly affect the G protein coupled amine receptor activity,cytokine receptor binding,antioxidant activity,neurotransmitter receptor activity,protease binding and other molecular functions related to play a therapeutic effect.And it mainly intermediary Interferon 17 signaling pathway,non-alcoholic fatty liver disease,TNF signaling pathway and other signaling pathways have been intervened.Youguiyin may mainly play a role in regulating lipid metabolism,liver drug enzymes,inflammation,osteoblast and osteoclastactivity treatment of GA-ONFH. |
