Empirical Study On Pathogenesis Of Youguiyin Preventing Steroid-induced Avascular Necrosis Of Femoral Head

OBJECTIVE To induce mice model of steroid-induced avascular necrosis of femoral head, observe changes of fatty substance metabolism, bone resorption and NO, and explore the pathogenesis of it.METHODS Forty wistar rats was randomly divided into four groups: normal control group ,model group,Youguiyin preventing group and Simvastatin preventing group. model group intraperitoneal injection of colibacillus endotoxin 20μg/kg per day two times, then gluteal injection of prednisolone per day three times,and intragastric administration with distilled water 10ml/kg per day; the model of youguiyin preventing group was induced like model group, but intragastricing administration with youguiyin 1.5g/kg per day; Simvastatin preventing group was induced like model group, but intragastricing administration with Simvastatin 2g/kg per day. After 6 weeks, measuring the content of NO, total cholesterol, triglyeride, blood phosphonium, blood calculus, ALP and trap-5b of venous blood got from the killed mice, and observeing histopathology change of the femur head,liver, hypothalamus, appendix cerebri and adrenal of the rats.RESULTS The model group comparing with the normal control group, Youguiyin preventing group, Simvastatin preventing group, the content of total cholesterol, triglyeride, ALP trap-5b, cortisol, acethropanand empty bone lacuna rate increased significantly (P<0.01), vitodynamics, cata-elasticity analysis, the content of blood phosphonium and calculus decreased significantly (P<0.01). Youguiyin preventing group and Simvastatin preventing group comparing with the normal control group, there are not statistical significance. CONCLUSIONS Rats model of steroid-induced avascular necrosis of femoral head can successfully induced through prednisolone and colibacillus endotoxin; The steroid-induced avascular necrosis of femoral head is complicated disease because of multiple factor interaceion,hormone restraining secretion of NO because of damaging blood vessel endothelium might be one of the pathogenesy of the steroid-induced avascular necrosis of femoral head;Youguiyin can prevent steroid-induced avascular necrosis of femoral head by regulating fatty metabolism, bone metabolism and NO.
