Objective:To observe the behaviors and lateral rein nucleus of electroacupuncture on the weight,water maze,open field experiment,sugar water consumption,etc.of depressed rats by electroacupuncture "Xie Yu Fang"(Baihui,Shenmen(bilateral),Taichong(bilateral))The content of ?CaMK?,GluR1 protein,IL-6,IL-10,IL-1?,NLRP3,IDO,5-HT serum content and gene expression were detected to explore the anti-depressive mechanism of electroacupuncture.Methods:Forty male SD rats screened by open field experiment were divided into four groups according to random number table: blank group,model group,sham acupuncture group and electroacupuncture group,10 rats in each group.Use restraint(3 h),clip tail(3min),heat bake(45-60 ?,5 min),fast and water(24 h),reverse day and night(24 h),swim in ice water at 4 ?(5 min),combined with orphanage and moist bedding to create a rat model of chronic unpredictable stress depression.Acupuncture once a day,normal feeding in the blank group without any treatment,the model group is fixed in the time period of acupuncture in the sham acupuncture group and the electroacupuncture group,5days is a course of treatment,two days between the two courses of treatment,a total of four A course of treatment.After 28 days,samples were taken for weight measurement,sugar water consumption test,water maze,and open field experiment.Western-blot and qPCR were used to detect ?CaMK?,GluR1 protein content and gene expression,qPCR brain protein IL-6,IL-10,IL-1?,NLRP3,IDO,5-HT gene expression,ELISA detection of serum IL-6,IL-10,IL-1?,NLRP3,IDO,5-HT content.Results : Compared with the blank group,the body weight,the number of horizontal exercises,the number of vertical exercises,and sugar water consumption of the model group all decreased(P <0.01),the difference was statistically significant;compared with the model group,the body weight,the number of horizontal exercises,and the number of vertical exercises of the electroacupuncture group,Sugar water consumption increased(P<0.01),the difference was statistically significant.Compared with the blank group,the space exploration percentage and space exploration distance percentage of the model group decreased(P <0.01),the difference was statistically significant;compared with the modelgroup,the space exploration percentage and space exploration distance percentage of the sham needle group and the electroacupuncture group increased.High(P <0.01),the difference was statistically significant;compared with the sham needle group,the percentage of space exploration and space exploration distance increased in the electroacupuncture group(P <0.01)),the difference was statistically significant.Compared with the blank group,the content of NLRP3,IL-1?,IL-6,IDO and gene expression of model group increased(P <0.01),while the content of IL-10,5-HT and gene expression decreased(P <0.01),?CaMK? protein content and gene expression increased(P <0.01),GluR1 protein content and gene expression decreased(P <0.01),the difference was statistically significant;compared with the model group,the sham acupuncture and electroacupuncture groups NLRP3,IL-1?,IL-6,IDO serum content and gene expression decreased(P <0.01),IL-10,5-HT serum content and gene expression increased(P <0.01),?CaMK? protein and Gene expression decreased(P <0.01),GluR1 protein and gene expression increased in the electroacupuncture group(P <0.01),the difference was statistically significant;compared with the sham acupuncture group,the serum content of NLRP3 and IL-6 in the electroacupuncture group and Gene expression decreased(P <0.01),IL-1? serum levels and gene expression decreased(P <0.05),the difference was statistically significant.Conclusions: 1.Electroacupuncture treatment for depression may be associated with down-regulation of protein content and gene expression of ?CaMK? and GluR1,which may produce anti-depressant effects,thereby alleviating depressive symptoms. 2.The anti-depression mechanism of electroacupuncture may be related to the fact that electroacupuncture can inhibit the inflammatory response induced by inflammatory factors.
