Background and objective: In 2018,there were 4.28 million new cancer cases in China and 2.86 million cancer deaths,accounting for 23.7%and 30.0% of the global cancer cases in the same year,respectively.The growth,infiltration and migration of tumor are Rel Ated to the disorder of coagulation and fibrinolysis system in vivo.However,most patients with malignant tumor are accompanied by abnormal functions of coagulation and fibrinolysis systems,which are often associated with acute and chronic arteriovenous thrombosis.The risk of venous thrombosis in patients with malignant tumor is about 7 times that of ordinary people.This tumor-Rel Ated event is known as Trousseau syndrome.Malignant tumors in traditional Chinese medicine belong to the category of "accumulation,syndrome",etc.,while the state of blood suppository and hypercoagulability of blood stasis syndrome belong to the category of "blood stasis syndrome" in Chinese medicine.The method of promoting blood circulation and removing blood stasis has certain research potential in preventing and treating hypercoagulability caused by chemotherapy.Taohongsiwu decoction(THSWD)originated from the qing dynasty Wu Qian works "Yi Zong Jin Jian",is the traditional Chinese medicine huoxue quyu classic.So can THSWD inhibit the development of tumors and in what ways?The research on network pharmacology is carried out in order to lay a foundation for the subsequent experimental research.Methods: Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)was used to screen the active components in six components of THSWD(Taoren,Honghua,Baishao,Chuanxiong,Shu Dihuang,and Danggui),and to search the active targets of all components meeting the screening conditions.OMIM and Gene Cards databases were used to search for prostate cancer and lung cancer disease Rel Ated targets.The network of "drug-ingredients-disease-targets" was established to obtain the key target of Taohongsiwu decoction in the treatment of prostate cancer and lung cancer.The protein-protein interaction(PPI)network of key targets was constructed,and Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were conducted on key targets using R software.Results: A total of 50 active ingredients were obtained,190 targets were found in the treatment of prostate cancer,and 198 targets were found in the treatment of lung cancer.PPI network analysis showed that the key targets with high critical degree in prostate cancer included ALB,IL-6,CASP3,EGFR,MAPK8,VEGFA,etc.The targets with high critical degree in lung cancer include JUN,AKT1,MAPK1,RELA,APP,IL-6,etc.Enrichment analysis of GO biological processes showed that prostate cancer involved 160 biological processes,among which the highly reliable biological functions and processes included nuclear receptor binding,transcription factor activity,steroid hormone receptor activity,DNA binding transcriptional activator activity,and proximal promoter sequence specific DNA binding.There are 169 biological processes involved in lung cancer,among which the biological functions and processes with high reliability include nuclear receptor activity,transcription factor activity,direct ligand regulation sequence specific DNA binding,protein heterodimer activity,steroid hormone receptor activity,heme binding,coenzyme factor binding and so on.KEGG enrichment analysis showed that there were 162 signaling pathways involved in prostate cancer,among which the most reliable ones were prostate cancer pathway,kaposi sarcoma-Rel Ated herpes virus infection pathway,fluid shear stress and atherosclerosis pathway,hepatitis b pathway,and human cytomegalovirus infection pathway.There are 108 pathways involved in lung cancer,among which the pathways with high reliability include age-rage signaling pathway,IL-17 signaling pathway,tumor necrosis factor signaling pathway,and endocrine resistance pathway,etc.Some pathways have been documented to play an important role in the occurrence and development of cancer.Conclusion: THSWD may have potential therapeutic effect on prostate cancer and lung cancer.The main components of the decoction include quercetin,luteolin,kaempferol,sterols,flavonoids,carotene and myricetin.THSWD may act on prostate cancer throughJUN,MAPK1,Rel A,AKT and other targets,as well as the prostate cancer pathway,IL-17 signaling pathway,fluid shear stress-Rel Ated pathway and other pathways.It may be that JUN,MAPK8,MAPK14,FOS and other targets,as well as IL-17 signaling pathway,tumor necrosis factor signaling pathway,age-rage signaling pathway and other pathways of diabetic complications have certain effects on lung cancer.
