At present,cancer seriously affects the health of people all over the world.In our country,lung cancer has the highest incidence rate among malignant tumors.Exploring the occurrence and development mechanism of lung cancer is one of the fields that urgently need to be studied.There are many targets and pathways related to the onset of lung cancer.Traditional Chinese medicine has its unique advantages in the treatment of lung cancer.This study screened out the high-frequency drug pairs for the treatment of lung cancer by Professor Piao Bingkui through outpatient prescriptions,and used the method of network pharmacology to explore the high-frequency drug pairs.The targets and mechanism of action for the treatment of lung cancer,and preliminary verification through cell experiments,in order to lay a certain foundation for further research on lung cancer mechanisms in the future.The research in this paper is mainly divided into the following parts:1.Screening of high-frequency drugs for the treatment of lung cancer by Professor Piao BingkuiObjective:To screen the high-frequency drugs used by Professor Piao Bingkui to treat lung cancer and to summarize Piao Lao’s experience in treating lung cancer.Method:Collect and sort out the outpatient prescriptions of Professor Piao Bingkui for the treatment of lung cancer,upload them to the integrated pharmacology platform and analyze them,and obtain high-frequency drug pairs for the treatment of lung cancer and potential targets for the treatment of lung cancer.Results:The high-frequency drugs screened in the outpatient prescriptions include Radix Platycodonis,Radix Radix Ophiopogon,Hawthorn,Shenqu-Malt,Radix Astragali-Tangerine Peel,Radix Astragali-Prince ginseng,etc.,which are all traditional Chinese medicines for strengthening the body.This is the same as Professor Piao Bingkui’s treatment.The academic thoughts of lung cancer are consistent.The high-frequency outpatient prescription targets include MAP4,MPO,ORM1,PTGS1,etc.The literature review is related to the tumor microenvironment and immunity,which further validates Professor Piao Bingkui’s righting thoughts.2.Research on the mechanism of high-frequency drugs on the treatment of lung cancer based on network pharmacologyObjective:To explore the target and mechanism of Radix Adenophora-platycodon on the treatment of lung cancerMethods:Firstly,the TCMIP database was used to collect the effective active chemical components of the"Adenophora-platycodon" drug pair,and to predict the target of the effective chemical components of the "Adenophora-platycodon" drug pair.Use Genecards,DisGeNET,OMIM,DrugBank and other databases to collect lung cancer targets.Then use Cytoscape3.5.1 to construct the target-compound network diagram,use the string database to construct the lung cancer-drug pair protein interaction network diagram,and analyze the network topology.Finally,use the DAVID online database to carry out GO function enrichment analysis and KEGG pathway enrichment analysis,to explore the relevant targets and signal pathways of"Adenophora-platycodon" medicine for the treatment of lung cancer.Results:The key compounds of the "Adenophora-platycodon" drug pair were selected as glycosides,coumarins,starch,triterpene acids,stigmasterol,phospholipids,amino acids,polysaccharides,oleanandane-type pentacyclic triterpene saponins,and flavonoids,Polyacetylenes,steroids,phenolic acids,fatty acids and other components,the key targets mainly include TGFBR2,TNF,ATP1A2,IL6,PTEN,etc.The key proteins of the PPI network of lung cancer-drug pairs include GAPDH,HSP90AA1,AKT1,PTEN,IL6,MAPK3,CASP3,ESR1,ACTB,TNF,etc.;the results of GO enrichment analysis show that "Adenophora-platycodon" has the main effect on local organisms Involved in protein kinase activity,nuclear receptor activity,oxidoreductase activity,kinase binding,transmembrane receptor protein kinase activity,lipid binding,exogenous transmembrane transport ATPase activity,nitric oxide synthase modulator activity,single Oxygenase activity,activin activated receptor activity,etc.The results of KEGG enrichment analysis showed that"Adenophora-platycodon" medicine mainly acts on prostate cancer,small cell lung cancer and other diseases,and the pathway with cancer,TGF-beta signaling pathway,adhesion junction,transcriptional disorders in cancer,PI3K-The Akt signal pathway,HIF-1 signal pathway,MAPK signal pathway and other signal transduction pathways are closely related.3.The effect of Adenophora-platycodon on the preparation of medicated serum and the effect of medicated serum on the proliferation of A549 cellsObjective:To prepare adenophora-platycodon drug-containing serum,and observe the effect of adenophora-platycodon drug-containing serum on the proliferation of lung adenocarcinoma A549 cells by MTT method.Methods:According to the experimental principles of serum pharmacology,the drug-containing serum of Adenophora Radix Platycodonis was prepared.The effect of the traditional Chinese medicine group and the control group on the proliferation of A549 cells was detected by MTT detection method after 24 hours.Results:Comparing each drug-containing serum group with the blank control group,the differences were statistically significant(P<0.05).Among them,the 10%group of traditional Chinese medicine has the highest cell inhibition rate of 60.3%.4.Scratch experiment to observe the effects of Adenophora Radix and Platycodon grandiflorum on the migration ability of A549 cellsObjective:To observe the effect of Adenophora Radix Platycodonis on the migration ability of A549 cells.Methods:The cell scratch test method was used to observe the effect of the drug-containing serum within 24 hours on the migration ability of lung adenocarcinoma A549 cells.Image J image software was used to analyze the area of cell scratches and calculate the percentage of cell scratch healing.Results:After 24 hours,the area of scratches between the drug-containing serum group and the blank control group changed significantly,and the percentage of scratches healed in the blank control group was greater than that of the traditional Chinese medicine group.Conclusion:1.Adenophora-platycodon drug pair is Professor Park Bing-kui’s high-frequency drug pair for the treatment of lung cancer.Professor Park Bing-kui’s high-frequency drug pair and medication for the treatment of lung cancer explain the academic thought of Professor Park Bing-kui’s treatment of lung cancer to strengthen the body and eliminate the evil.2.Adenophora-platycodon is the result of multiple pathways and multiple targets for the treatment of lung cancer.This study uses network pharmacology to explain the possible related targets and signal pathways for the occurrence and development of lung cancer,and to explore its mechanism in the future Lay a certain theoretical foundation.3.Adenophora-platycodon can inhibit the proliferation and migration of lung adenocarcinoma cells to a certain extent on the drug-containing serum,but at present,it is still necessary to conduct in-depth research on protein and related conduction pathways.
