Objective:To investigate the molecular mechanism of Sculellaria barbata combine with Hedyotis diffusa in the treatment of lung metastases and bone metastases from breast cancer by using network pharmacology and GEO chip difference analysis.Methods:Using R language difference Analysis GEO Chip,searching TCMSP,Pub Chem and Swiss Target Prediction databases to screen the targets of Sculellaria barbata and Hedyotis diffusa,searching String,Hiplot databases to enrichment analysis PPI,GO and KEGG of the differential targets of Sculellaria barbata combine with Hedyotis diffusa in the treatment of lung metastases and bone metastases from breast cancer.Results:Baicalein,luteolin and quercetin are the main active ingredients of the medicine pair of Sculellaria barbata and Hedyotis diffusa.They can target proteins such as VEGFA,SMAD4 and MMP9 in lung metastases from breast cancer or MMP9,MMP2 and CCNB1 in bone metastases,regulating the interaction of proteins such as AKT1,MYC and CCND1 in lung metastases from breast cancer or MMP2,CCNB1 and PTGS2 in bone metastases.In the cell components,such as transcription regulator complex,transferase complex in lung metastases from breast cancer or transferase complex,intrinsic component of presynaptic membrane in bone metastases,they can activate molecular function such as beta-catenin binding,alcohol dehydrogenase activity in lung metastases from breast cancer or nuclear receptor activity,ligand-activated transcription factor activity in bone metastases,regulating biological process such as regulation of cell cycle phase transition,regulation of Wnt signaling pathway in lung metastases from breast cancer or hormone metabolic process,steroid metabolic process and response to oxygen levels in bone metastases.activating signal pathway such as Wnt,TGF-? in lung metastases from breast cancer or P53,Micro RNA,PI3K-AKt in bone metastases.Conclusion:In this study,we constructed the network of Sculellaria barbata and Hedyotis diffusa-lung metastases and bone metastases from breast cancer-Target point interaction network,and concluded from the genetic and molecular point of view that the mechanism of Sculellaria barbata and Hedyotis diffusa in the treatment of lung metastases and bone metastases from breast cancer is related to the network regulation of multi-active components,targets,cellular components,molecular functions,biological processes and pathways.
