ObjectiveBased on the network pharmacology,the potential mechanism of Jianpi Lishi Huayu formula in the treatment of prostate cancer was discussed,and the network of"drug composition target action pathway"was constructed.Animal experiments were carried out to verify the mechanism of Jianpi Lishi Huayu Recipe in the treatment of prostate cancer through multiple targets and channels.Methods1 Network pharmacology researchThrough six databases of TCMSP,OMIM,TTD and disgenet,the related targets of prostate cancer were searched and selected.Based on the same database of TCMSP and TCMID,the related compounds and corresponding targets of single drug in"Jianpi Lishi Huayu Recipe"were searched and collected,and the network of drug active component corresponding targets was drawn.The intersection of drug targets and disease targets was drawn by Venny2.1 online software By making Wayne map,the protein protein interaction network(PPI)network was constructed with the common targets,and the topological structure value of PPI network was calculated through Cytoscape to complete the network visualization analysis.Finally,the key target proteins were analyzed by KEGG enrichment pathway analysis and go analysis through David database to draw the"drug component target pathway"network of Jianpi Lishi Huayu formula.2 Experimental research36 nude mice were randomly divided into 6 groups(6 rats in each group):model group(tumor bearing control),western medicine group(ENDOSTAR),"Jianpi Lishi Huayu formula"whole formula group,Junyao group(Astragalus,liquorice),chenyao group(Acanthopanax senticosus,Psoralea)and adjuvant group(turmeric,wangbuluoxing).The PC3prostate cancer bearing nude mice model was established in each group.When the tumor volume reached about 50mm~3,The mice in the model group were given distilled water and the mice in the experimental drug group were given intraperitoneal injection and gavage of corresponding concentration twice a day for 14 days.After 14 days,the mice were killed,the eyeballs were removed,the tumor bodies were taken and weighed,the tumor weight and tumor inhibition rate were compared in each group;the levels of VEGF and e NOS in serum were detected by ELISA;Western The protein expression levels of phosphatidylcholine3-hydroxykinase(PI3K),serine threonine protein kinase(Akt),p-Akt and e NOS in tumor tissues were detected by blotting.Results1 results of network pharmacology analysisThere are 938 prostate cancer related targets were selected by tcmsp and other six databases.Based on tcmsp and tcmid databases,113 active components of single drug in Jianpi Lishi Huayu Recipe were searched and collected,and 209 targets were in total.There were 70 targets of Jianpi Lishi Huayu Recipe for prostate cancer,and 69 targets were found to be able to interact with each other by protein protein interaction network diagram analysis Through go analysis and KEGG enrichment pathway analysis,it is concluded that Jianpi Lishi Huayu Recipe can treat prostate cancer by promoting apoptosis,inhibiting cell proliferation,cell migration and invasion,and inhibiting tumor angiogenesis.2 experimental researchJianpi Lishi Huayu Recipe group and its disassembled prescriptions can inhibit the tumor growth of PC3 prostate cancer bearing mice,at the same time,they can reduce the content of VEGF and e NOS in mice serum,which has a significant difference compared with the model group(P<0.05).The whole recipe group and its disassembled prescriptions group can reduce the expression of PI3K,p-Akt and other protein in PI3K/Akt related pathway key points,among which the highest inhibition rate is in the whole recipe group The adjuvant group was the best group to inhibit the growth of prostate cancer PC3.ConclusionsJianpi Lishi Huayu Recipe group and its disassembled prescriptions group have a certain anti-tumor effect on PC3 prostate cancer bearing mice,which can inhibit tumor angiogenesis.Its treatment mechanism may be through blocking PI3K/Akt pathway conduction and the expression of related site proteins,reducing the content of VEGF and e NOS in mice serum,so as to play the role of inhibiting tumor angiogenesis and blocking tumor growth The results are also consistent with the results of previous network pharmacology...
