Deer skin glue is prepared by decocting the skins of deer animals such as Cervus nipponT.or Cervus elaphus L.and has the functions of enriching blood,invigorating qi and tonifying kidney-yin.At present,the main problems are that the raw materials of deerskin glue are difficult to identify,the decocting process,the pharmacological action and the quality standard research work are not perfect.In view of the above problems,this paper uses DNA barcode technology to identify and distinguish deer skins of different deer species,and establishes a new identification method.At the same time,the boiling process of deerskin glue was improved.In addition,the immunomodulatory effect of deerskin glue was deeply studied.The specific research results are as follows:1.DNA barcode identification of deer skin species and authenticityBased on COI barcode sequence of samples,NJ tree was established to analyze the difference between genuine deerskin and adulterated products.NJ tree results show that genuine sika deer skin and red deer skin are gathered into independent branches with approval rate of100%,showing obvious monocline.Samples of adulterated products also gathered into separate ones,with approval rating of 100%.This study proves that DNA barcode technology based on COI sequence can effectively identify the authenticity of deer skin.2.Study on boiling technology of deerskin glueTaking gum yield,total protein content and comprehensive score as the main indexes,through orthogonal experiment optimization,through visual analysis and variance analysis,we can see that the change of decocting times?factor B?level has a significant impact on the experimental results.According to the actual production needs,the boiling conditions are adjusted to A2B3C1 that is,adding 10 times of water,decocting 5 times,2 hours each time,that is,decocting 5 times,adding 1 time.0 times the amount of water,2 hours each time.3.Verify the immunoregulatory effect of deerskin glueThe immune hypofunction model of mice was established by intraperitoneal injection of cyclophosphamide?Cy?method,and the immunoregulation effect of deerskin glue was studied.The results showed that deerskin glue could promote the organ index and spleen lymphocyte proliferation of hypoimmunity mice,enhance the energy metabolism level of abdominal cavity?M Phi?of mice and the ability to phagocytize neutral red,increase the number of CD4+,CD8+cells in external week blood,and the ratio of CD4+/CD8+.Experiments proved that deerskin glue has certain immunoregulation effect.4.Establishment of quality standard for deerskin glueAccording to the detection indexes under donkey-hide gelatin in the 2015 edition of pharmacopoeia of the People's Republic of China,we have determined the moisture,heavy metals,insoluble sludge and amino acid contents of 10 batches of self-made deerskin glue samples.Based on the detection results of 10 batches of samples,the temporary deerskin glue content is not more than 15%;The content of Pb is not more than 1.36mg/kg,the content of Cd is not more than 0.1mg/kg,the content of As is not more than 0.1mg/kg,the content of Hg is not more than 0.1mg/kg,and the content of Cu is not more than 0.83 mg/kg.Insoluble sludge content does not exceed 2%;The total content of amino acids is not less than 80%.
