The main pharmacologically active components of medicinal plants are usually secondary metabolites,and the secondary metabolites are the products of plants adapting to the environment.In addition to the genetic factors,the secondary metabolites of medicinal plants are regulated by genetic factors,and the ecological factors such as geographical environment and climate conditions It has an important impact on it,that is,the environment leads to different quality of the same kind of medicinal materials.The wild 'Di Wu' resources are widely distributed in China,and there are bound to be significant differences in the quality of traditional Chinese medicines from different regions.The accurate identification of 'Di Wu' medicinal materials and the selection of high-quality medicinal materials are important for their rational development and clinical application.The comprehensive quality evaluation system of the medicinal materials from the place of origin has not yet been established.This study completed the collection of wild resources from 16 places in 8 provinces based on the provenances of 'Di Wu' produced in China.According to traditional Chinese medicine identification methods such as character identification,microscopic identification and physical and chemical identification.The 'Di Wu' samples were identified,and the identification results were consistent with the characteristics of genuine 'Di Wu' medicinal materials recorded in the literature and in the “Hubei Province Traditional Chinese Medicine Quality Standards”(2018 edition).At the same time,a rapid identification system of 'Di Wu' medicinal materials was successfully constructed based on the DNA barcode ITS2 sequence.The traditional identification technology combined with the DNA barcode identification technology of 'Di Wu' medicinal material combined identification system ensured the accuracy of 'Di Wu' medicinal material identification results and laid a foundation for its clinical safety.The compound identification system constructed in this study ensured that the origin of the 'Di Wu' medicinal material collected was accurate.On this basis,five pharmacologically active ingredients(anhuienoside E,glycoside St-14 a,hemsgiganoside B,hederasaponin B)of 16 places of origin were completed using HPLC.Analysis of differences found that the content of anhuienoside E and hemsgiganoside B in 'Di Wu' samples from Duzhenwan,Changyang,Hubei was significantly higher than other origins(P <0.05).Saponin glycoside St-14 a was highest in Bijie,Guizhou,Banqiao,Enshi,Hubei and Yuanbao Enshi,Hubei 'Di Wu' samples(P <0.05).The contents of saponin flacidoside ? in Banqiao,Enshi 'Di Wu' samples from Hubei were significantly higher than those from other origin samples(P <0.05),and the contents of saponin hederasaponin B was most hightest in Banqiao,Enshi and Baiyiping,Wufeng,Hubei.At the same time,the sum of the contents of the five detected components in the sample of Banqiao,Enshi in Hubei was the highest.Cluster analysis based on chemical composition showed that 'Di Wu' in Hubei had certain similarities in chemical composition and content with 'Di Wu' medicinal materials in Chongqing,Anhui and Guizhou.Ultraviolet spectrophotometry was used to determine the total saponins content of the Chinese medicinal materials from 16 places of origin,and the results showed that the total saponins content of the Chinese medicinal materials from different places was significantly different.Among them,'Di Wu' from Bijie,Guizhou had the highest total saponins content(P <0.05),followed by Banqiao,Enshi in Hubei,Xuxian in Anhui,and Sangzhi in Hunan.'Di Wu' from Shennongjia in Hubei,and Hengshan,Hunan had the lowest contents.In this study,a compound identification system for traditional identification and molecular identification of 'Di Wu' medicinal materials were established to ensure that the basis of 'Di Wu' medicinal materials was accurate.At the same time,the content of 5 saponins and total saponins in 16 different origins were measured,and the content and total saponin content of 'Di Wu' medicinal materials were used as the quality evaluation indicators.The differences,and thus determine the origin of high-quality 'Di Wu' medicinal materials.The above research lays the foundation for the selection of 'Di Wu' medicinal materials and clinical medicine,and at the same time provides certain data support for the selection of high-quality varieties of 'Di Wu' in the later stage.
