Objective: To study the synergistic effect of Radix Angelicae Sinensis(RASI)combined with hormone in the treatment of bronchial asthma,and to explore its mechanism of action from the perspective of HMGB1/TLR4/NF-κB signaling pathway.At the same time,to investigate the alleviation effect of RASI on the yin deficiency symptoms caused by hormone treating asthma.Methods: Using OVA to replicate the asthma model by Sensitization and stimulation,we observed the effects of RASI,hormone and their combination on asthma behavior,lung function and lung histopathology.Determining the contents of TNF-α,IL-1β and IL-6 in BALF and expression levels of key proteins and genes of HMGB1/TLR4/NF-κB signaling pathway in lung tissue,the goal of the experiment is to study the synergistic effect and mechanism of RASI combined with hormone in the treatment of bronchial asthma.At the same time,on the basis of replicating asthma model,the body weight,food and water intake,voluntary activities of asthmatic mice were determined,discussing the alleviation effect of RASI on the yin-deficiency symptoms caused by hormone treating asthma.All the experimental datas were analyzed by SPSS 24.0 software,and experimental results are expressed in the form of “ ±s”.When P<0.05 or P<0.01,the comparisons among groups were considered to be statistically significant.Results: 8g/kg RASI,0.05g/kg hydrocortisone sodium succinate(HSS)and their combination can significantly alleviate asthma attacks and their symptoms,improve lung function and pathological changes of lung tissue,reduce the contents of TNF-α,IL-1β and IL-6 in BALF,and inhibit the expressions of key proteins and genes of HMGB1/TLR4/NF-κB signaling pathway.the effect of their combination on the above indicators generally increased further,and some indicators have statistical significance(P<0.05 or P<0.01).Meanwhile,after the combination of RASI and hormone,RASI can significantly improve the symptoms of yin-deficiency such as increased intake and drinking water,decreased body weight,and increased autonomic activity in mice induced with hormone.Conclusions: RASI and its combination have obvious antiasthmatic effect.After combination,its antiasthmatic effect is further enhanced,and inhibiting HMGB1/TLR4/NF-κB signaling pathway is one of the mechanisms of anti-asthmatic action.At the same time,RASI can alleviate the yin-deficiency symptoms caused by hormones. |
