Objective:The aim of this research is to study the therapeutic effect of Radix Angelicae Sinensis(RADA)on Yin-deficiency asthma and its effect on mucus hypersecretion,explore the molecular mechanism of RADA in the intervention intervention of airway mucus hypersecretion in Yin-deficiency asthma based on IL-13-mediated "crosstalk" between STAT6 and AQP5.Methods:The mice were randomly divided nto control group,model group,ambroxol group and RADA high,medium and low doses(n = 12).The Yin-deficiency asthma model was reproduced with OVA and thyroid tablets to observe the effects of asthma symptoms,lung function,pulmonary index,lung fluid clearance function,pulmonary histopathology.To determine total airway mucus solids and Muc5 ac content,airway goblet cell hyperplasia and lung tissue Muc5 ac expression levels.To determine serum related active substances(including IgE,IL 13,TNF-?,etc.)and the expression levels of STAT6,AQP5 and NF-?B in lung tissue,to study the effect of RADA on Yin-deficiency asthma,hypersecretion of airway mucus in Yin-deficiency asthma and its molecular mechanism.Results:RADA can significantly relieve the symptoms of asthma in mice,improve lung function,reduce lung index and lung tissue wet-dry weight ratio,relieve pulmonary histopathological changes.RADA can reduce the total solids and Muc5 ac content in BALF of Yin-deficiency asthma of mice,inhibit airway goblet cell proliferation reduces the expression of Muc5 ac in lung tissue.RADA can reduce the levels of serum IgE,IL-13 and TNF-? in Yin-deficiency asthma of mice,inhibit the expression of STAT6 and NF-?B in lung tissue,and promote the expression of AQP5 in lung tissue;at the same time,correlation analysis showed that IL-13 was positively correlated with STAT6.Conclusions:RADA has a significant therapeutic effect on Yin-deficiency asthma,which can inhibit airway goblet cell hyperplasia and Muc5 ac expression,and relieve Yin-deficiency asthma mucus hypersecretion.Among them,RADA through crosstalk between IL-13 and STAT6 and then inhibited the expression of STAT6 and promoted the expression of AQP5 are one of the mechanisms for relieving Yin-deficiency asthma and relieving the hypersecretion of Yin-deficiency asthma airway mucus.
