Objective: To study the intervention effect of Proanthocyanidins from Lycium ruthenicum,on the aging skin model of mice induced by ultraviolet(UVA+UVB)irradiation combined with subcutaneous injection of D-galactose.The relative expressions of matrix metalloproteinase-1(MMP-1),matrix metalloproteinase-3(MMP-3)and epidermal growth factor receptor(EGFR)in skin tissue of mice were determined by RT-q PCR.The effect of vegetable on aging skin in mice. Methods: 50 healthy SPF Kunming female mice were randomly divided into 5 groups: 10 in normal control group(A),10 in model group(B),and 10 in positive control group(C).10 in low-dose of proanthocyanidins from Lycium ruthenicum Group(D),10 in high-dose of proanthocyanidins from Lycium ruthenicum Group(E);establishe the mouse aging model;RT-q PCR was used to determine the m RNA expression levels of MMP-1,MMP-3 and EGFR in skin tissues of each group. Results: Compared with the model group, the relative expression of MMP-1,MMP-3 and EGFR in the skin tissue of the normal control group was significantly decreased, the difference was statistically significant(p<0.05);compared with the model group, the expression of MMP-1,MMP-3 and EGFR in the skin tissue of mice in the positive control group and the low-dose group of proanthocyanidins from Lycium ruthenicum decreased significantly, the difference was statistically significant(p<0.05),and the low dose of proanthocyanidins in the Lycium ruthenicum, the relative expression of the group was lower than that of the positive control group, and the difference was statistically significant(p<0.05);the relative expression of MMP-1,MMP-3 and EGFR in skin tissue of mice in the high-dose group of Lycium barbarum proanthocyanidins was significantly lower than that in the low-dose group, the difference was statistically significant(p<0.05).Conclusion: Lycium ruthenicum proanthocyanidins can reduce the expression levels of MMP-1,MMP-3 and EGFR in aging skin tissues of mice, and the reduction level is greater than that of the same dose of vitamin E intervention. It shows that Lycium ruthenicum proanthocyanidins is a strong antioxidant and has certain effect in delaying skin aging.
