Objective: To investigate the effects of proanthocyanidins from Lycium ruthenicum Murr on the skin structure of mouse aging model.Method:A total of 50 Kunming female rats of Specific Pathogen Free(SPF)were selected.50 mice were randomly divided into 5 groups,10 in each group.The grouping was: normal control group(group A,10),model group(group B,10),positive control group(group C,10),low-dose group(group D,10),high-dose group(group E,10).The mouse skin aging model was established by subcutaneous D-galactose injection combined with long-wave black spot effect ultraviolet(UVA)and medium-wave erythema effect ultraviolet(UVB)irradiation.The purpose of the treatment of drugs using proanthocyanidins from Lycium ruthenicum Murr.Except for group A experimental mice,the other groups were injected with 5% D-galactose(10 ml/Kg)subcutaneously on the back of the neck,and each group was irradiated with UV(UVA+UVB)on the skin of the neck and back every other day.The number of days was 40 days,thereby establishing a mouse skin aging model.The drug intervention started on the 11 th day of modeling.Group C was given vitamin E(VE)solution 50 mg/Kg,group D was given low dose drug 50 mg/Kg,group E was given high dose drug 100 mg/Kg for 30 days.The histopathological study method was used to detect the changes of skin epidermis stratum corneum,dermal layer thickness and collagen fiber in each group;the relative expression of type I and type III collagen mRNA was detected by RT-qPCR.Results: Compared with group A,the degree of keratinization of the skin epidermis in the neck and back of the mice in group B was observed to increase,the thickness of dermis,collagen fibers and the relative expression of type I collagen mRNA decreased,and the relative expression of type III protein mRNA increased.(P < 0.05).Compared with group B,it was observed that group C,group D and group E had decreased skin thickness of the epidermis of the neck and back,and dermal layer thickness,collagen fiber and the relative expression of type I collagen mRNA increased.The relative expression of type III collagen mRNA was decreased,and the change of group D was more significant than that of group C,and the change of group E was more significant than that of group D(P<0.05).Conclusion: Proanthocyanidins from Lycium ruthenicum Murr have anti-aging effects by repairing the skin epidermis stratum corneum and dermis collagen fibers of mouse skin aging model.
