Objective To research the effect of the Proanthocyanidins in Lycium ruthenicum on oxidative damage and the contents of caspase-9 and caspase-3 of mice after subcutaneous injection of D-galactose combined with ultraviolet(UVA+UVB)radiation.Methods Fifty SPF female KM rats were randomly divided into 5 groups : normal control group(CON,10rats),aging model group(MOL,10rats),vitamin E group(VE,10rats),low concentration of proanthocyanidins in Lycium ruthenicum group(LPC,10rats),high concentration of proanthocyanidins in Lycium ruthenicum group(HPC,10rats).Except for the CON group,each group was given subcutaneous injection of 5% D-galactose(5ml/kg)daily,and given ultraviolet UV irradiation every other day,modeling for 40 days continuously to establish the aging model.When the 11 th of modeling,the group VE received vitamin E(50 mg/kg),group LPC was given PC(50 mg/kg),group HPC was given PC of Lycium barbarum(100 mg/kg),Continuous gavage of stomach for 30 days.Observing the state of the mice in each group and the appearance of the skin and recoring the weight changes of the mice during the modeling period.After the mold is finished,score on the skin of the back of the neck,take off the skin about the back and neck,measured the water content of the skin,enzyme biochemical method measurie SOD activity?GSH-PX content?MDA content?Hyp content;measuring the content of caspase-9 and caspase-3 with ELISA.Result After using D-galactose combined with ultraviolet(UVA + UVB)radiation deal with the mouse,in the group MOL,the skin of the neck on the back of the mouse appeared wrinkle deepening,desiccative in part,desquamation,erythema,color sink and so on appears,weight growth was slow,Compared with the group MOL,The symptoms of group VE,group LPC and group HPC were significantly reduced.The activity of SOD and the content of water content of skin and GSH-PX and Hyp in aging group MOL were reduced,the content of MDA caspase-9 and caspase-3 were increased(P<0.05);Contrast with the group MOL,the activity of SOD and the content of water content of skin and GSH-PX and Hyp in group VE and group LPC and HPC were increased,the content of MDA and caspase-9 and caspase-3 were reduced(P<0.05);And comparing LPC or HPC with the group VE,the difference between the two groups was statistically significant(P<0.05).Conculution the proanthocyanidins in Lycium ruthenicum can inhibition the oxidative damage of D-galactose combined with ultraviolet(UVA + UVB)radiation,and down-regulation of the expression level of caspase-9 and caspase-3,inhibit apoptosis,delay senescence of skin in a certain degree,and the effect is better than vitamin E,and have concentration dependence.
