Objective: To explore the expression of USP2 in hepatocellular carcinoma and its biological role.Methods: The expression of USP2 in hepatocellular carcinoma and its adjacent tissues was detected by Western blotting;The transcription of USP2 mRNA and the expression of USP2 in hepatocellular carcinoma cell lines HepG2,SK-Hep1,SMMC-7721 and normal hepatocyte line L02 were compared by ways of qRT-PCR and Western blotting;HepG2cells were randomly divided into experimental group,negative control group,blank group,the experimental group was transfected with shRNA(Oligo).Negative control group was transfected with negative control nucleotide sequence,except of blank group.Transfection effect was tested by qRT-PCR and Western blot;CCK-8 test and cell clone formation test were used to detect the proliferation of HepG2 cells in the transfection group and the control group.Transwell test and Matrigel invasion test were used to detect the changes of metastasis and invasion ability of HepG2 cells in the transfection group and the control group respectively.Results: The expression of USP2 in hepatocellular carcinoma tissues was higher than that in adjacent tissues(P <0.05);The expression of USP2 protein and the transcription of USP2 mRNA in three hepatocellular carcinoma cell lines were all higher than that in normal hepatocytes L02(P <0.01);The expression of USP2 protein and the transcription of USP2 mRNA in transfected HepG2 experimental group was higher than that in negative control group and blank group(P <0.01).CCK-8:HepG2 cell lines with high expression USP2 have significantly increased the proliferation ability of lower expression cell lines(P<0.01);Cell cloning experiment: The proliferation ability of HepG2 cell lines with low expression USP2 was significantly lower than that of HepG2 cell lines with high expression USP2(P <0.01);Transwell test: the migration ability of HepG2 cell lines with high expression of USP2 was higher than that of HepG2 cell lines with low expression ofUSP2(P <0.01).Matrigel test: the invasion ability of HepG2 cell lines with high expression of USP2 was higher than that of HepG2 cell lines with low expression of USP2(P <0.01).Conclusion: The high expression of USP2 in hepatocellular carcinoma can promote hepatocellular carcinoma cells proliferation,metastasis and invasion,which may provide a new target and theoretical basis for the diagnosis and molecular targeted therapy of hepatocellular carcinoma.
