Objective:to study and explore the silence Ebpl HepG2 liver cancer cell proliferation,the influence of the ability of invasion and metastasis,for the treatment of tumor and prognosis provide an accurate prediction,and to provide new ideas for the selection of tumor targeting drug targets.Methods:1.Immunohistochemical staining was used to detect the expression of Ebpl protein in 60 liver cancer tissues and adjacent tissues.2.Western blot method was used to detect the expression of Ebpl protein in hepatocellular carcinoma cell lines(Hep G2,Hep 3B,Alexander,Huh7,sk-hep 1),and finally selected Hep G2 as the lentivirus transfection cell.3.The expression and localization of GFP-Ebp1 protein in HepG2 hepatocellular carcinoma cell lines were observed under inverted fluorescence microscope.4.Using lentivirus mediated shma-ebpl to infect HepG2 hepatocellular carcinoma cells with puromycin to screen for metastatic cells.5.Western blot was used to detect the silencing effect of Ebp1 in the shma-ebp1 group and the negative control group.6.Cell proliferation of positive shma-ebpl group,blank control group and empty plasmid group were detected by cck-8 method7.The effect of Transwell invasion assay on the invasion and migration of HepG2 in hepatocellular carcinoma cells after silent Ebp1 gene was detectedResults:1.Ebpl protein was expressed in high expression in liver cancer tissues,and low or no expression was found in adjacent tissues and normal tissues.2.Ebp1 protein is highly expressed in liver cancer cell lines,and the expression is highest in HepG2 hepatocellular carcinoma cells.3.GFP-Ebp1 can be expressed in HepG2 hepatocellular carcinoma cells,and the expression site is mainly located in the cytoplasm,with no or no expression in the nucleus.4.Western blot showed that the expression of Ebp1 protein was inhibited in shma-eb41 group compared with the control group.5.CCK8 experiment showed that the cell proliferation of shma-ebp1 group decreased significantly(P<0.05)compared with the blank control group and the negative control group.6.Transwell experiment showed that the cell migration and invasion ability of shma-ebp1 group were decreased compared with that of the empty plasmid group(P<0.05).Conclusion:The expression of slow virus targeting to silent Ebpl can reduce its invasion ability,and this finding will provide new ideas for finding molecular targets to inhibit the metastasis of liver cancer.
