Effect Of Shenyan Xiaobai Granules On Expression Of Podocalyxin And ZO-1 In Rat With Diabetic Nephropathy

Objective : To observe the effect of Shenyan Xiaobai granules on urinary albumin(UA),expression of prodocyte-relating proteins podocalyxin and ZO-1in rats model with diabetic nephropathy(DN),and to investigate the mechanism of Shenyan Xiaobai granules in the prevention and treatment effect of DN.Methods: After unilateral nephrectomizing,we gave the models of rats of diabetic nephropathy with High Fat-High Sucrose Diet and Streptozotocin with single intraperitoneal injection,then separate the rats into 5 groups:the blank control group,the model contrast group,the group with high dosage of Shenyan Xiaobai granules,the group with low dosage of Shenyan Xiaobai granules and the group with Benazepril Hydrochloride(Lotensin).After gavage for 8 weeks,test the urea nitrogen values.Color the Pathological Sections with the PAS staining,using light microscope to observe the change of rat kidney tissue pathological;test the expression level of podocalyxin and ZO-1 with immunohistoche mistry method.Results: Shenyan Xiaobai granules can significantly reduce the urinary protein excretion,the group with Shenyan Xiaobai granules is much obvious than the model group(P<0.05);compared with the the group with Benazepril Hydrochloride,the group with low dosage of shenyanxiaobai granules treatment is more significant(P<0.05);it can lighten the Mesangial Proliferation,Basement Membrane incrassation and the fuse of foot process;Compared with the model group,the protein expression level of podocalyxin and ZO-1 in the other groups increased(P<0.05).Conclusion:Shenyan Xiaobai granules can decrease the UAE of the model rats of diabetic nephropathy,through the increasing of the expression of proteins on the podocyte podocalyxin and ZO-1,to reduce the discharging of the Proteinuria,to reach the purpose of protecting kidney,delay the progress of diabeticnephropathy in model rats.
