Objective:To investigate the intervention mechanism and prevention effect of this prescription on coronary heart disease rats with myocardial infarction,and to provide a theoretical basis for the clinical application of Zhuangtongyin in the treatment of myocardial infarction in coronary heart disease.Through the observation of Zhuangtongyin in the coronary heart disease rats with TNF-?,IL-6,IL-10,NE,REN,AngII,ALD,ET-1,TGF-?1 levels,at the same time,it provides research ideas or strategies for the development of Guangxi Zhuang-medicine with the characteristics of myocardial infarction in coronary heart disease.Methods:SPF SD rats were randomly divided into blank group,sham operation group,model group,Zhuangtongyin?low,middle,high?dose group and positive drug control group,a total of 7 groups.The model group,Zhuangtongyin?low,medium,high?dose group and positive drug control group were surgically ligated to the left anterior descending branch of the left coronary artery to establish a myocardial infarction model of coronary heart disease.The sham operation group only opened the chest without ligation.The Zhuangtongyin group was given intragastric administration according to low?6.8g/kg?,middle?13.6g/kg?and high?27.2g/kg?doses,the positive drugs were administered with Compound Danshen Dripping Pills?80 mg/kg?after modeling,serum and myocardium were taken after 4 weeks of administration.?1?The death situation and general condition of the rats were observed and recorded in the process of drug delivery.?2?Heart rate changes were analyzed by electrocardiogram.?3?Myocardial pathological changes were observed by HE staining.?4?The levels of serum TNF-?,IL-6,IL-10,REN,AngII,ALD,NE and ET-1 were detected by enzyme linked adsorption assay?ELISA?.?5?The expression of ET-1 protein in the myocardium of Western blot was expressed by the method of Western blot.?6?The expression of TGF-?1 in myocardial tissue was detected by immunohistochemistry?IHC?and fluorescence real-time quantitative?RT-PCR?method.The data were expressed as"mean±standard deviation?x±s?".SPSS21.0 statistical software was used to analyze the data between groups using one-way analysis of variance?ANOVA?,P<0.05 was considered statistically significant.Results:?1?Comparison of the general situation:The rats in the model group showed a marked decrease in diet,poor spirit,less activity,dry coat,dullness,obvious weight loss,and rapid breathing,etc.After drug intervention,ZTY?low,medium,and high?dose groups,The above-mentioned phenomena in the positive drug control group were improved to varying degrees,and the ZTY low,middle dose group,and positive drug control group showed relatively obvious improvement.?2?Heart rate changes:The heart rate of rats in the model group was significant higher,and the heart rate was significantly improved after drug intervention.There was no significant difference between groups?P>0.05?.?3?HE staining:Inflammatory cell infiltration,myocardial fibrosis and collagen deposition were obvious in the model group,after the treatment of drugs,the above phenomena in the ZTY?low,middle,high?dose group and the positive drug control group all improved obviously,and the effect of middle dose group was obvious.?4?Enzyme-linked adsorption?ELISA?:Compared with the blank group and the sham operation group,the level of serum TNF-?,IL-6,IL-10,REN,AngII,ALD,NE and ET-1 in the model group increased significantly?P<0.05?;Compared with the model group,the levels of TNF-?,IL-6,REN,AngII,ALD,NE and ET-1 in ZTY?low,middle and high-dose?group and positive drug control group were significant decreased?P<0.05?,and IL-10 significant increased?P<0.05?,the difference was statistically significant;Compared with low dose group,the levels of TNF-?,IL-6,REN,AngII,ALD,NE and ET-1 in middle,high and positive drug control group were significant decreased?P<0.05?,the difference was statistically significant,There was no significant change in the expression of IL-10?P>0.05?,and there was no significant difference in the difference;The changes of TNF-?,IL-6,IL-10,REN,AngII,ALD,NE and ET-1 were not significant?P>0.05?,but the difference was not statistically significant.?5?Western blot:The relative expression of ET-1 protein in the model group was significant higher than that in the blank group and the sham operation group,after the treatment of drugs,the ZTY?low,middle and high dose?group and the positive drug control group were all decreased,and the decrease of the middle dose group was relatively obvious.?6?TGF-?1 immunohistochemistry?IHC?:The model group showed strong positive performance.After drug intervention,ZTY?low,medium,high?dose group and positive control group showed positive performance.TGF-?1 measured by RT-PCR:Compared with the blank group and the sham operation group,the level of TGF-?1 in the myocardial tissue of the model group was significant increased?P<0.05?,and the difference was statistically significant;Compared with the model group,the level of TGF-?1 in the ZTY?low,middle,high dose?group and the positive drug control group decreased significantly?P<0.05?,and the difference was statistically significant;Compared with the low dose group,the level of TGF-?1 in the middle,high and positive control groups was significant decreased?P<0.05?,and the difference was statistically significant;There was no significant difference in TGF-?1 between the middle,high and positive control groups?P>0.05?.Conclusion:?1?Zhuangtongyin can improve the general condition of rats,reduce inflammatory cell aggregation,inflammation factors?TNF-??IL-6?ET-1?TGF-?1?and neurohumoral factors?NE?REN?AngII?ALD?related to the release,there are some therapeutic effect on myocardial ischemia with coronary heart disease.?2?The mechanism of Zhuangtongyin for the treatment of myocardial ischemia in rats with coronary heart disease,may be correlated with the decreased accumulation of inflammatory cells,inflammatory?TNF-??IL-6?ET-1?TGF-?1?and humoral factors?NE?REN?AngII?ALD?released or interfere with the synthesis,and the effect of middle dose group was the best.
