Objective:To observe the effects of Zhuang Tong Yin(ZTY)on the Mechanism of oxidative stress,SOD,MDA,GSH-PX and ET-1 and NO in the rats with myocardial ischemia of coronary heart disease and blood stasis syndrome,To explore the protective effect and mechanism of this side on myocardium in rats with myocardial ischemia of coronary heart disease and blood stasis syndrome,For the development of Guangxi specific Zhuang medicine to provide coronary heart disease to provide relevant scientific basis.Methods:SPF grade Wistar rats were randomly divided into seven groups:blank control group,sham operation group,model group,positive control group,ZTY low,middle and high dose group,The model group,positive control group,ZTY low,middle and high dose group were treated by direct coronary artery anterior descending coronary artery occlusion method to establish coronary heart disease model of myocardial ischemia and blood stasis,Sham operation group only thoracotomy without ligation of left coronary artery,The intragastric dose of the ZTY groups were 6.8 g·kg-1·d-1for the low dose group,13.6g·kg-1·d-1 for the medium dose group and 27.2g·kg-1·d-1 for the high dose group,The positive control group was treated with compound Danshen Dripping Pill 80mg·kg-1·d-1,the indexes were measured at the end of the fourth week of administration.The histopathological changes of myocardium were observed by ordinary light microscope;The levels of serum SOD,GSH-PX,MDA and NO were measured by UV-Vis spectrophotometer,the content of SOD and MDA in myocardium,Serum ET-1 levels were measured by microplate reader.The data obtained from the experiment were expressed as mean ± standard deviation(± s).The difference between the two groups was statistically analyzed by statistical analysis.P <0.05 was considered statistically significant.Results:(1)Behavioral observation,the experimental rats with myocardial ischemia of coronary heart disease and Blood Stasis model with apathetic,yellow hair,slightly worse color,claw color darker,lips and tongue dark,less moving Baotuan,diet and other common blood stasis Performance,positive control group and Zhuang Tong drink each dose group of rats were improved to varying degrees.(2)Electrocardiogram detection,The rats in the model group were treated with left coronary artery for 10 to 30 minutes.The electrocardiogram II lead appeared in the S-T segment elevation of more than0.2mv,An electrocardiogram was performed one week after surgery,to I,II,av L lead abnormal Q wave for the model to establish a successful sign.(3)Pathomorphology of ordinary light microscopy,Under HE staining,compared with the model group,the pathological changes of myocardial tissue in the treatment group were reduced to varying degrees.(4)Oxidative stress test,Compared with the model group,the contents of SOD,GSH-PX,NO and SOD in the serum of the middle and high dose group rats were significantly higher than those in the model group,and the levels of MDA,ET-1 and MDA in serum were decreased in middle and high dose groups(P<0.05).Compared with the positive control group,the majority of the high dose group did not show significant difference(P>0.05).Conclusions:(1)The results of the above test indicators suggest that ZTY can reduce the degree of myocardial injury in rats,delay the progress of myocardial ischemia of coronary heart disease,on myocardial ischemia of coronary heart disease have a certain effect and And the effect of ZTY high dose group was slightly better than that of low and middle dose group.(2)The results of this study suggest that the effect of Zhuangtongyin on coronary heart disease with myocardial ischemia and blood stasis syndrome is the result of overall regulation under the influence of various factors,and its mechanism of action may be related to the increase of antioxidant enzymes SOD,GSH-PX and vasodilatation factor NO,decrease the levels of MDA and vasoconstrictor factor ET-1,improve oxidative stress and reduce the degree of myocardial ischemic injury in rats.
