Objective:To investigate the interventive effects of Zhuang Tong Yin(ZTY)on the expression of AngⅡ,AT-1R and Cx43 in rats with MIRI(myocardial ischemia-reperfusion in Jury).Methods:One hundred SPF grade SD rats were divided into NS group,sham-operated group(thread without ligation),model group,ZTY low do-se group(6.8g·kg-1),ZTY middle dose group(13.6g·kg-1),ZTY high dose group(27.2g·kg-1)and Compound Salvia Pellet dose group(0.08g·kg-1).Administrated with drug for 4 weeks,through ligation the left anterior d-escending coronary artery 30 min,again perfused for 60 min to observe E-CG and put down myocardial infarct proportion,RT-PCR method to de-tect the myocardial tissue in the expression of Ang Ⅱ,AT-1R and Cx43,Western Blot method to detect the protein expressions of AT-1R and Cx43 in the myocardial tissue.Results:(1)ZTY low dose group,middle dose group and high dose group can reduce myocardial ischemia reperfusion arrhythmia of rats(P ﹤ 0.05).(2)RT-PCR:Compared with the NS group group,the myocardial ischemia-reperfusion in Jury rats,ZTY low dose group,ZTY middle dose group,ZTY high dose group and Compound Salvia Pellet dose group myocardial tissue AngⅡand AT-1R increased significantly(P﹤0.05),Cx43 reduced(P﹤0.05).Compared with ischemia-reperfusion injury rats,ZTY low dose group,ZTY middle dose group,ZTY high dose group and Compound Salvia Pellet dose group myocardial tissue AngⅡand AT-1R reduced significantly(P﹤0.05),Cx43 increased(P﹤0.05),ZTY middle dose group better than those of other groups(P﹤0.05).There was no significant difference between ZTY each dose group and Compound Salvia Pellet dose group in AngⅡ,AT-1R and Cx43(P﹥0.05).(3)Western Blot:Compared with the NS group group,the myocardial ischemia-reperfusion in Jury rats,ZTY low dose group,ZTY middle dose group,ZTY high dose group and Compound Salvia Pellet dose group myocardial tissue AT-1R is increased significantly(P﹤0.05),Cx43 reduced(P﹤0.05).Compared with ischemia-reperfusion in Jury rats,ZTY low dose group,ZTY middle dose group,ZTY high dose group and Compound Salvia Pellet dose group myocardial tissue AT-1R reduced significantly(P ﹤0.05),Cx43 increased(P﹤0.05),ZTY middle dose group better than those other groups(P﹤0.05).There was no significant difference between ZTY each dose group and Compound Salvia Pellet dose group in AT-1R and Cx43(P﹥0.05).(4)ELISA:Compared with the NS group group,the myocardial ischemia-reperfusion in Jury rats,ZTY low dose group,ZTY middle dose group,ZTY high dose group and Compound Salvia Pellet dose group myocardial tissue AngⅡincreased significantly(P﹤0.05).Conclusion:ZTY could reduce myocardial ischemia-reperfusion arrhythmia and have protective effects on myocardial ischemia-reperfusion.ZTY could reduce the expression of Ang and inhibit ATⅡ-1R,then regulate the expression of Cx43 in the myocardial tissue,which is more significant in the middle dose group than those in the low dose group and high dose group.
