Significance Of Abnormal Expression Of MicroRNA-575 In Recurrent Abortion

Objective : 1.To explore miRNA-575 expression in recurrent spontaneous abortion(RSA)patients and the normal early pregnant women.To investigate the differential expression of Vascular Endothelial Growth Factor(VEGF)in recurrent spontaneous abortion patients and normal early pregnant women.To analyze the expression of miRNA-575 and VEGF in recurrent spontaneous abortion patients and normal early pregnant women,and to explore the relationship between miRNA-575 and VEGF and their pathogenesis in patients with recurrent spontaneous abortion.The mechanism in the process.Methods:1.Real-time PCR was used to detect the expression of miRNA-575 in the villus tissues of 28 patients with recurrent spontaneous abortion(experimental group)and the same number of normal pregnant women(control group)who met the inclusion criteria information.2.Real-time quantitative PCR and Western Blot were used to detect the expression of vascular endothelial growth factor(VEGF)in recurrent abortion patients and normal early pregnant women.3.To observe the relationship between miRNA-575,VEGF and recurrent spontaneous abortion and the correlation between miRNA-575 and VEGF expression.All data were statistically analyzed using SPSS 23.0 statistical software.Results: The results of RT-PCR showed that the expression level of miR-575 was 4.92�5.14 in the villus tissues of patients with recurrent spontaneous abortion in the experimental group,while the expression level of miR-575 was 1.49�1.35 in the normal villus tissues of pregnant women in the control group.It can be seen that the expression level of miR-575 in the experimental group was significantly higher than that in the normal control group(P<0.001),and the difference was statistically significant.2.The results of qRT-PCR showed that the expression level of VEGF in the villus tissues of the experimental group was 0.42�0.43,while in the control group,the expression level of VEGF was 1.45�1.13,and the RSA group had villi.Theexpression level of VEGF in tissues was significantly lower than that in normal pregnant women(P<0.005).At the same time,Western Blot showed that the expression of VEGF protein in the villus tissue of the experimental group was significantly lower than that of the control group.3.Correlation analysis of miR-575 and VEGF expression in RSA group Pearson linear correlation analysis showed that miR-575 was negatively correlated with VEGF expression in villi tissues of experimental group(? = 0.802),and normal pregnant women in the control group were female villi.There was also a negative correlation between miR-575 and VEGF expression in tissues(? = 0.695).Conclusion:1.The expression of miRNA-575 in the villus tissues of patients with recurrent spontaneous abortion was higher than that of normal pregnant women,suggesting that miRNA-575 may be involved in the occurrence of recurrent spontaneous abortion.2.The insufficient increase of VEGF expression in villus tissue may be one of the important factors in the occurrence of recurrent spontaneous abortion.3.The overexpression of miRNA-575 may be related to the inhibition of VEGF expression,which may lead to vascular dysplasia,which may be related to RSA.
