OBJECTIVE To investigate the expression of LPA2 and hes-1 in 67 cases of moderately differentiated gastric adenocarcinoma and the expression levels of mRNA and histone in 10 cases of freshly differentiated gastric cancer were analyzed,providing a theoretical basis for the exploration of new molecular pathological diagnostic markers for gastric cancer in the future.METHODS(1)67 cases of moderately differentiated gastric adenocarcinoma tissues and their corresponding paracancerous tissues were collected after gastric cancer resection and confirmed by the pathological diagnosis.Tissue fixation,dehydration,paraffin impregnation,paraffin embedding,continuous section,HE staining and immunohistochemical staining were performed.(2)10 fresh surgical gastric cancer specimens and the corresponding paracancerous tissues were collected and stored in RNAlater solution and tissue protein preservation solution.Subsequently,pathological diagnosis confirmed that the specimens were moderately differentiated gastric adenocarcinoma.(3)Immunohistochemical S-P method was used to detect the expression of LPA2 and Hes-1 in 67 cases of moderately differentiated gastric adenocarcinoma and adjacent tissues.(4)QRT-PCR was used to detect the relative expressions of LPA2 and Hes-1 in moderately differentiated gastric adenocarcinoma tissues and adjacent tissues at the mRNA level in 10 cases.(5)Western Blot analysis the relative expression of LPA2 and Hes-1 in moderately differentiated gastric adenocarcinoma tissues and adjacent tissues of 10 cases.(6)Calculated Spearman correlation coefficient and analyzed the correlation between the expression of LPA2 and Hes-1 in the tissues of 67 cases of moderately differentiated gastric adenocarcinoma.RESULTS(1)The positive high expression rate of LPA2 in 67 cases of moderately differentiated gastric adenocarcinoma.was 73.1%,which was significantly higher than that in paracancerous tissue(p <0.01).(2)In 67 cases of moderately differentiated gastric adenocarcinoma,the high expression rate of Hes-1 was 79.1%,which was significantly higher than the corresponding paracancerous tissue,with statistical significance(p < 0.01).(3)Through the analysis of clinical and pathological features,LPA2 was not related to age,gender,tumor diameter and other factors of patients withmoderately differentiated gastric adenocarcinoma(p > 0.05),but related to lymph node metastasis and invasion depth of tumor(p < 0.05).(4)Hes-1 was not related to patient age,gender,tumor size and other factors(p > 0.05),but was related to lymph node metastasis and infiltration depth in moderately differentiated gastric adenocarcinoma(p < 0.05).(5)By calculating Spearman correlation coefficient,the positive correlation between the expressions of LPA2 and hes-1 in the tissues of moderately differentiated gastric adenocarcinoma can be obtained(r=0.673,p < 0.05).(6)The mRNA and protein expression levels of 10 cases of moderately differentiated gastric adenocarcinoma and adjacent tissues were analyzed.The expression levels of LPA2 and hes-1 in moderately differentiated gastric adenocarcinoma were significantly higher than those in adjacent tissues(p < 0.05),which had statistical significance.CONCLUSION This study confirmed that the high expressions of LPA2 and Hes-1 are highly expressed in moderately differentiated gastric adenocarcinoma tissues,and the expressions level of LPA2 and Hes-1 mRNAs and proteins support the immunohistochemical results.Therefore,the combined application of LPA2 and Hes-1 can be used as potential markers for the molecular pathological diagnosis of gastric cancer and the prognosis of gastric cancer patients in the future,and provide new targets for the molecular treatment of gastric cancer.
