OBJECTIVE To study the LPA2 affection on the proliferation,migration and invasion ability of gastric cancer cell,and whether there is correlation among LPA2 and Notch1 signal and curcumin.METHODS(1)By si-994、si-1098、si-1146 and Negtive Control(NC)four siRNAs transfection and pcDNA3.1-LPA2 plasmid transfection in SGC-7901,Western blotting was used to measure the expression of LPA2.(2)Western blotting was used to measure the expression of E-cadherin and Vimentin.(3)The change of migration and invasion ability was tested by transwell experiment.(4)MTS assay was used to measure the proliferation of SGC-7901,Used the flow cytometry to test the effect on tumor cell apoptosis.(5)After the interference of SGC-7901 with siRNA,the expression of Notch1 was detected by Western blotting.(6)The SGC-7901 was tranfected by siRNA,after each group were treated with 0,10,and 20μmol/L curcumin,the change of migration ability was tested by transwell.The above experiments were repeated three times.RESULTS(1)si-1098 and si-1146 were successful in knockout LPA2,E-cadherin expression increased,Vimentin expression decreased,and si-1098 was followed up by comprehensive comparison.PcDNA3.1-LPA2 plasmid transfected SGC-7901 cells successfully,E-cadherin expression decreased and Vimentin expression increased.(2)Compared with group NC,the invasion and migration ability of siRNA group was reduced;The overexpression group had higher invasion and migration ability than the 0 μg group.(3)Compared with group NC,siRNA had low proliferative ability and high apoptosis ability.Compared with the 0 μg group,the overexpression group had higher proliferative ability and lower apoptotic ability.(4)The expression of Notch1 in group NC was lower than that in group siRNA.(5)The invasion and migration ability of cells transfected with LPA2 0,10,20μmol/L curcumin decreased gradually.CONCLUSION(1)These data indicate that expression of LPA2 regulates themigration and invasion ability in gastric cancer cell.(2)There was a positive correlation between LPA2 protein and Notch1 protein expression in SGC-7901 gastric cancer cells.(3)curcumin may inhibit the migration of SGC-7901 cells by LPA2.
