Objective: By examining the genetic polymorphisms of MMP-2,MMP-7 and MMP-9 and their expression levels in serum,to exploring the association between MMP-2,MMP-7 and MMP-9 gene polymorphism and premature rupture of membranes in Tibetan and Han nationality in Qinghai area.In order to provide a basis for the etiology of premature rupture of membranes and guides the prevention of premature rupture of membranes.Methods:(1)From January 2016 to December 2017,randomly selected women who were hospitalized in the obstetrics department of the affiliated hospital of Qinghai university,Qinghai Red Cross hospital and Qinghai provincial people's hospital,then randomly divided those 408 women into two groups,one group is normal pregnancy and delivery,the other group is premature rupture of membranes before delivery.Using PCR-RFLP,DNA sequencing and SNP typing techniques to analyze the polymorphisms of MMP-2,MMP-7 and MMP-9 in Tibetan and Han nationality normal pregnancy and fetal membranes.(2)The expression levels of MMP-2,-7 and-9 in serum of normal pregnancy and early fetal membrane failure were detected by ELISA.(3)Data analysis was performed by relative statistical methods and draw a conclusion.Results:(1)Only one genotype of MMP-2 rs243865 and MMP-2 rs2285053 was detected in fetal membrane tissues of both Tibetan and Han nationality,and the expression level of MMP-2 in serum of both Tibetan and Han nationality was not detected.(2)The allele frequency and genotype frequency of MMP-7 rs11568818 were significantly different between the premature rupture of membranes and the normal pregnancy group(P<0.01).Allele G was one of the risk factors for premature rupture of membrane(OR>1,95% CI lower limit>1),however,there was no significant difference in allele frequency and genotype between the Tibetan and Han groups and between the normal pregnancy groups(P>0.05).The expression level of MMP-7 in Tibetan and Han nationality serum was not detected.(3)The genotype and allele frequency of MMP-9 rs3918242 were not significantly different between the normal pregnancy group and the premature rupture of membranes(P>0.05),and there was no significant difference in allele frequencies and genotypes between the premature rupture of membranes and the normal pregnancy group in Tibetan and Han nationality groups(P>0.05).The expression level of MMP-9 in serum of the same nationality was statistically significant between the premature rupture of membranes group and the normal pregnancy group(P<0.05),but there was no significant difference between the premature rupture of membranes and normal pregnancy groups in Tibetan and Han nationality groups(P> 0.05).Conclusions:(1)MMP-2 rs243865 and MMP-2 rs2285053 only got one genotype in the fetal membrane tissues of Tibetan and Han nationality in Qinghai region;the expression level of MMP-2 in serum of Tibetan and Han women was not detected,it may be related to its extremely low content or small amount of samples and experimental methods,which need to be further explored.(2)The genetic polymorphism of MMP-7 rs11568818 may be related to the susceptibility of premature rupture of membranes in Tibetan and Han nationality in Qinghai.Allele G is one of the risk factors for premature rupture of membranes,but there was no significant difference in gene polymorphism between Tibetan and Han nationality in Qinghai.The expression level of MMP-7 in serum is not detected too.Considering that it may be related to its extremely low content or small amount of samples and experimental methods,which need to be further explored.(3)The genetic polymorphism of MMP-9 rs3918242 may be unrelated to the susceptibility to premature rupture of membranes in Tibetan and Han nationality in Qinghai,and it is not significant between Tibetan and Han nationality in Qinghai.The high expression of MMP-9 in serum of pregnant women with premature rupture of membranes may be related to the occurrence of premature rupture of membranes in Tibetan and Han nationality in Qinghai,but there is no difference between Tibetan and Han nationality.
