Objective: To investigate the association between MMP-1?-3?-8 gene polymorphous and premature rupture of membranes in Tibetan and Han nationalities of Qinghai at genetic level,and to investigate the association between MMP-1?-3?-8 express in serum and premature rupture of membranes in Tibetan and Han nationalities of Qinghai on molecular level,analyze the relationship between the polymorphism and expression of MMP-1?-3?-8 and premature rupture of membranes,in order to provide some experimental basis for the etiology study and prevention of premature rupture of membranes.Methods:390 cases of fetal membrane tissues and 120 cases of serum samples of normal pregnant and premature rupture of membranes women in Tibetan and Han were collected from the affiliated Hospital of Qinghai University,the people's Hospital of Qinghai Province and the Red Cross Hospital of Qinghai Province from December 2015 to October 2017.PCR and gene sequencing technology were used to analysis polymorphism and genotype of MMP-1rs1799750 ? MMP-3rs617819 ?MMP-8rs11225395,the MMP-1?-3?-8 in serum was detected by enzyme-linked immunosorbent assay(Elisa).Finally,the statistical method is used to analyze the data.Results: The genotypes and alleles of MMP-1rs1799750 were significantly different between premature rupture group and control group in the same nation(P < 0.05),and the risk of PROM in pregnant women with allele G was decreased;The genotypes and alleles of MMP-3rs617819 were not significantly different between premature rupture group and control group in the same nation(P>0.05).The genotypes of MMP-8rs11225395 were significantly different between premature rupture group and control group in the same nation(P < 0.05),but there was no significant difference in alleles(P>0.05).There was no significant difference in genotype distribution and allele of MMP-1rs1799750?MMP-3rs617819?MMP-8rs11225395 between the Han and Tibetan premature rupture of membranes group,as well as Han and Tibetan control group(P >0.05).The expression of MMP-1-3-8 in serum were significantly different between premature rupture of membranes group and the control group in the same nation(P < 0.05).Conclusions:The gene polymorphisms of MMP-1rs1799750 and MMP-8rs11225395 may be related to the susceptibility to PROM in Tibetan and Han nation in Qinghai area.The risk of PROM in Tibetan and Han women with allele G of MMP-1rs1799750 may be lower in Qinghai.The gene polymorphisms of MMP-3rs617819 may not be related to the susceptibility to PROM in Tibetan and Han nation in Qinghai area.The high expression of MMP-1?-3?-8 in serum may be related to the occurrence of PROM in Tibetan and Han nation in Qinghai area.MMP-1rs1799750?MMP-3rs617819?MMP-8rs11225395 gene polymorphism and the expression of MMP-1?-3?-8 in serum were not significantly different between Tibetan and Han nation in Qinghai area.
