Objective: To analyze the characteristics of MMP-2?-7?-8?-9 gene polymorphism in the fetal membrane tissue of puerpera and detect the expression of MMP-2?-7?-8?-9 in serumin in Hui and Han nationalities of Qinghai area.And analyze the relationship between the gene polymorphism and expression of MMP-2?-7?-8?-9 and premature rupture of membranes,In order to provide experimental basis for early diagnosis,prevention and treatment of premature rupture of membranes.Methods: Collect 387 cases of fetal membrane tissue and 120 cases of serum samples of the pregnant women from Affiliated hospital of Qinghai university,Qinghai provincial people's hospital and Qinghai Red Cross hospital from December 2015 to October 2017.PCR and gene sequencing technology were used to analysis gene polymorphism,at the same time,to test the expression levels of MMP-2?-7?-8?-9 in the serum through ELISA,Finally,the statistical method is used to analyze the data.Results: The genotype frequency and allele frequency of MMP-7rs11568818 were significantly different between the normal group and premature rupture of membranes group in the same nation(P<0.05),pregnant women carrying the allele of G are more likely to suffer from premature rupture of the membranes;The genotype frequency of MMP-8rs11225395 were significantly different between the normal group and premature rupture of membranes group in the same nation(P<0.05),but there was no significant difference in allele frequency(P>0.05);The genotype frequency and allele frequency of MMP-9 rs3918242 were not significantly different between premature rupture group and control group in the same nation(P>0.05);The genotype frequency and allele frequency of the above three locus were not different between the two groups of different nationalities(P>0.05).The expression of MMP-8 and MMP-9 in serum were significantly different between the normal group and premature rupture of membranes group in the same nation(P<0.05),but not different in different nations(P>0.05);The expression of MMP-2 ?-7 was not detected in serum.Conclusions: The gene polymorphisms of MMP-7rs11568818 may be related to the susceptibility to the premature rupture of membranes in Hui and Han nationalities in Qinghai area,pregnant women carrying the allele G may be more likely to suffer from premature rupture of the membranes;The gene polymorphisms of MMP-8 rs11225395 may be related to the susceptibility to the premature rupture of membranes in Hui and Han nationalities in Qinghai area;The gene polymorphisms of MMP-9 rs3918242 might not be related to the susceptibility to premature rupture of membranes in Hui and Han nationalities in Qinghai area.The gene polymorphisms of MMP-7 rs11568818,MMP-8 rs11225395 and MMP-9 rs3918242 are not different between the Han and Hui nationality.The high expression of MMP-8 and MMP-9 in serum may be related to the occurrence of the premature rupture of membranes in the Hui and Han nationalities in Qinghai area(P<0.05),and were not different in Hui and Han nationalities(P>0.05).
