Objective: To investigate the correlation of rs671 polymorphism of ALDH2 gene of the essential hypertension in Maotai,Guizhou and their alcohol drinking habit.Methods: With permanent residents(residence time of no less than 5 years)between 35 and 75 years old in Maotai town as the object of study,the existing resident health management archives in Maotai sub-district and community health service station were analyzed,and the resident health information was obtained by door-to-door questionnaire survey and return visit on phone.After the preliminary questionnaire survey,blood specimens from 300 people were collected using random stratified sampling method,and the number of blood specimens actually collected was278,among which 198 ones belong to hypertension group and 80 ones belong to non-hypertension group(non-hypertensive people).A record of the study objects in terms of their age,gender,habit of alcohol consumption,habit of physical exercise,history if illness and history of medicine-taking were made,together with the measurement of their body weight,height,blood pressure and heart rate and the calculation for their physical fitness index.Collection of all blood specimens was arranged for morning,and the objects of study were clearly informed that eating and drinking was forbidden during the 8 hours before the blood collection and all drugs had better be taken after the blood collection except for certain ones that must be taken on time.Determination of biochemical indexes such as uric acid,homocysteine,aspartate Aminotransferase,alanine Aminotransferase,glucose,total cholesterol,triglycerides,high density lipoprotein,low density lipoprotein cholesterol,glomerular filtration rate and glycated hemoglobin,as well as the genotyping of rs671 site of ALDH2 gene from 278 DNA samples based on KASP technique,were altogether done by the testing center of Third Affiliated Hospital of Zunyi Medical University.All data obtained were analyzed by SPSS 19.0 software,and the group representation of samples with genotype of rs671 site of ALDH2 gene was tested using H-W balance.The multi-factor Logistic regression model was used to effectively evaluate the correlations between various factors and analyze the influencing factors.Results: 1.The rs671 genotype of ALDH2 gene accorded with H-W balance in hypertension group and non-hypertension group.2.The GG genotype frequencies of rs671 of ALDH2 gene were 24.1% and 75.9% in hypertension group and non-hypertension group,The GA/AA genotype frequencies of rs671 of ALDH2 gene were 41.3% and 58.7% in hypertension group and non-hypertension group,respectively,with a significant difference in rs671 genotype frequency of ALDH2 gene between the two groups(P<0.05).Besides,the G allele frequencies of the two groups were 26.4% and 73.6%,the A allele frequencies of the two groups were 42.7%and 57.3%,respectively,with a significant difference in rs671 genotype and allele frequency of ALDH2 gene between the two groups(P<0.05).3.In terms of alcohol consumption,age,physical fitness index,UA,GFR,Hcy and HbA1 c,t-test and chi-square test results revealed that there was significant difference in the comparison between hypertension group and non-hypertension group.However,no significant difference was detected concerning gender,AST,ALT,GLU,CHOL,TG,HDL-C,LDL-C,no history of hyperlipidemia or diabetes mellitus between hypertension group and non-hypertension group(P>0.05).4.With the status of hypertension as dependent variable,multivariate logistic regression analysis was applied to analyze the variables with statistical significance in the above analysis.The results showed that the risk factors related to hypertension were mutant genotype of rs671 of ALDH2 gene,alcohol consumption,age,physical fitness index and HbA1c(P<0.05),while no obvious effect of UA,GFR and Hcy level was found on hypertension statistically(P>0.05).5.In male patients without hypertension,there was evident difference in age difference among different alcohol consumption groups(P<0.05).In male patients with hypertension,there were evident difference in physical fitness index,UA and TG difference among different alcohol consumption groups(P<0.05),but there was no significant difference in other indicators among the three groups(P>0.05).Therefore,in male patients with essential hypertension,there was evident difference in mutant genotype of rs671 of ALDH2 gene difference among different alcohol consumption groups(P>0.05).6.There was no correlation between alcohol consumption and mutant genotype of rs671 in male patients without hypertension with no adjustment or with factor adjustment such as physical fitness index,age,GFR,Hcy andHbA1c(P>0.05).Conclusion: 1.There is no obvious correlation between the GA/AA genotype of rs671 of ALDH2 gene and alcohol consumption in male patients with essential hypertension in urban areas of Maotai Town,Guizhou Province;2.The GA/AA genotype of rs671 of ALDH2 gene is a risk factor for essential hypertension in urban residents of Maotai Town,Guizhou Province;3.Age and drinking are risk factors for essential hypertension in urban residents of Maotai Town,Guizhou Province.
