Objective: The aim of this study is to estimate the association between SELEgene Polymorphism and essential hypertension susceptibility and provideevidence on the effect of the SELE gene on the hypertension.Methods: The association between E-selectin gene polymorphism andessential hypertension was examined in458patients and571healthy subjects,The classical risk factors were recorded in both Patient and control groups,such as body weight, height, smoking, alcohol consumption and so on.Phenol-Chloroform method was used to extract genomic DNA. Genotypes ofSELE gene polymorphism was detected using Matrix-assisted laser desorptionionization time-of-flight mass spectrometry technology (MALDI-TOF-MS).Measurement data were compared with group t test; count data were comparedwith x2test; Odds ratio (OR) and95%confidence interval (CI) were obtainedfrom unconditional univariate and multivariable logistic regression model toevaluate associations between SELE polymorphism and essential hypertensionrisk. Hardy-Weinberg genetic equilibrium (HWE)、LD analysis、haplotypeanalysis were tested using SHEsis software (http://analysis2.bio-x.cn/SHEsisMain.htm). MDR method was used to test the introaction between SNP.All testswere two-sided with α is0.05. Results:⑴Baseline characteristics:There were statistically differences in BMI, TG,smoking and alcohol consumption between cases and controls respectively(P<0.01).⑵Association between SELE polymorphism and essential hypertension:FourSNPs genotype frequency distributions were in agreement with the HWEin controls (P>0.05). The rs4786genotype and allele frequencies were st-atistically different between cases and controls (P<0.05). There was signif-icant association between rs4786polymorphism and essential hypertension(adjusted Avs.G:OR=0.800,95%CI=0.667-0.960;AAvs.GG:OR=0.645,95%CI=0.446-0.934;GA+AAvs.GG: OR=0.757,95%CI=0.579-0.990),after adju-sted with the factors of age, sex, BMI, smoking and drinking. Therewas no significant association between rs5355、rs5359、rs5368Polymorphi-sm and essential hypertension risk.⑶Subgroup analysis was performed by the gender:In the male group, there was significant association between rs4786polymorphism and essentialhypertension genotype(Avs.G:OR=0.618,95%CI=0.466-0.820;GAvs.GG:OR=0.440,95%CI=0.280-0.694;AAvs.GG:OR=0.459,95%CI=0.267-0.789;GA+AAvs.GG:OR=0.447,95%CI=0.294-0.679), but in the female group,therewas no significant association between the rs4786polymorphism and the r-isk of essential hypertension. For rs5355,rs5359,rs5368,there were nosignificant association of essential hypertension when subgroup analysis bythe sex.⑷interaction analysis: The MDR analysis suggested that there were nointeraction between four SNPs in relation to essential hypertension. However, logistic regression models found that rs4786polymorphism gene-genderinteractions were statistically significant associated with essential hypertension.⑸haplotype analysis:The A-C-A-C haplotype of rs4786、rs5355、rs5359andrs5368has a protective effect on essential hypertension (OR=0.085,95%CI=0.674-0.962,P=0.017). The other haplotypes were not statisticallysignificant associated with essential hypertension (P>0.050).Conclusion: The rs4786polymorphism was associated with essentialhypertension; rs4786polymorphism may be a independent risk factor ofessential hypertension in male population; The A-C-A-C haplotype may play arole to decrease the risk of essential hypertension.
