With the increasing incidence of allergic rhinitis(AR),allergic diseases represented by asthma and allergic rhinitis have received increasing attention as global public health issues.Currently,drug treatment for AR can only temporarily relieve symptoms,and cannot fundamentally treat allergic rhinitis.Immunotherapy is currently the only treatment that can change the process of antigen-host immune response through mechanisms,Therefore,it is the only hope of eradicating AR.Histamine is an endogenous amine in the central and surrounding tissues.It plays a role in the initial stage of allergy.The binding of receptors to histamine can cause inflammation.At present,clinically used antihistamines are mainly histamine receptor-1(HR1)antagonists,which can only be symptomatic treatment.Clinical studies have shown that this method does not have a significant clinical effect on patients with allergic rhinitis.Histamine Receptor-4(HR4)is a newly discovered histamine receptor that involves all immune cells of the AR pathological immune response and has been shown to mediate immune regulation,if it can inhibit the regulation of HR4,The preparation of histamine receptor-4 antagonists can play a regulatory role in the development of AR.Therefore,in this experiment,we used the histamine receptor-4 peptidomimetic-liposome-CTB vaccine which had been prepared in the previous period.Through constructing the AR guinea pig model,we observed the behavior of the experimental group and counted the guinea pigs.Subsequently,relevant samples were collected and processed.ELISA was used to detect changes in the expression levels of factors known to be associated with AR.The results were compared by ELISA.Control group(ie,AR group)IL-6,VEGF,HIF-1α,CCL-22.The expression of CCL-17 was significantly increased and the expression of IL-2 was significantly decreased.The expression of IL-6,VEGF,HIF-1α,CCL-22,and CCL-17 were decreased in the PT1 and PT2 vaccine-administered groups.The expression level of-2 increased significantly,and both were close to the normal level,indicating that the PT1 and PT2 vaccines have a certain inhibitory effect on AR,that is,the histamine receptor-4 mimic peptide has inhibitory effect on AR,among which the PT1 vaccine group is closer In the normal group,the effect of PT1 vaccine was better.The innovation of this project lies in the use of histamine receptor antagonists as the research idea of this subject.The prepared HR4 epitope mimic peptide vaccine has the advantages of simple preparation and low cost;it greatly reduces the dosage and time;the property is stable,without Endotoxin and source of infection,small side effects and other advantages.Secondly,guinea pigs were selected as animal models in this study.Due to their relatively large size,they were relatively resistant to the attack of toxic components of the antigen and were easier to obtain at the detection stage.
