Objective: To investigate the expression of SHIP2 in epithelial ovarian cancer and its relationship with clinicopathological features。Materials and Methods: From January 2012 to June 2018,paraffin sections of ovarian tissues(100 cases of benign ovarian tissues and 100 cases of epithelial malignant tumors)were selected from gynecological department of Yuhuangding Hospital,Yantai,affiliated to Qingdao University.Immunohistochemical technique(IHC)was used to detect the location and intensity of SHIP2 protein expression in these tissues.Fresh ovarian tissue specimens(30 benign ovarian tissues and 30 epithelial ovarian malignant tumors)from December 2016 to June 2018 were collected.,the expression of SHIP2 in epithelial ovarian malignant tumors and benign ovarian lesions was assessed by RT-PCR,and the relationship between the expression of SHIP2 protein and gene and the clinicopathological characteristics of epithelial ovarian cancer was discussed.Result:(1)The positive expression rate of SHIP2 in epithelial ovarian malignant tumors was68% by immunohistochemical method,which was significantly higher than that in benign ovarian lesions(21%).The difference was statistically significant(P < 0.001),and the expression of SHIP2 was mainly in the cytoplasm.The expression of SHIP2 was significantly correlated with FIGO staging,differentiation and lymph node metastasis,with P values of 0.001,0.010 and 0.00,respectively.But there was no significant correlation with age,P=0.748.(2)The relative expression of SHIP2 in epithelial malignant ovarian tumors was2.479 times higher than that in benign ovarian lesions(P=0.008)detected by RT-PCR.The relative expression of SHIP2 in advanced,poorly differentiated and lymph node metastasis groups was 3.107,2.164 and 1.687 times higher than that in early,highly differentiated and non-lymph node metastasis groups,respectively,P=0.008.The expression of SHIP2 was 0.004,0.022 and 0.028 in different age groups,but the relative expression of SHIP2 in the younger group was 0.656 times higher than that in the older group,and P= 0.054.There was no difference in the expression of SHIP2 in the younger group.Conclusion:(1)The expression of SHIP2 in epithelial ovarian malignant tissues was significantly higher than that in benign ovarian lesions,which suggested that SHIP2 might play an important role in the pathogenesis of epithelial ovarian cancer.(2)SHIP2 may play an important role in the occurrence and progression of epithelial ovarian cancer.SHIP2 may become an effective biological molecule for early detection,diagnosis,prognosis and treatment of epithelial ovarian cancer.
