Objective:To discuss the protein and mRNA expressions of NPR-A,P38 MAPK and Cytoskeleton Tubulin-? in gastric cancer and normal gastric tissue and explore the clinical significance and the interactions of them.Which provide a new ideal for the study of pathogenesis of gastric cancer and a potential drug target for targeted therapy.Method:We has collected 41 cases of the gastric cancer and 41 cases of normal gastric tissues that 10 cm away from cancer edge in the Affiliated Hospital of Chengde Medical University from September 2016 to September 2017,There were 27 males and 14 females,aged from 38 to 85 years old,17 cases with high-grade differentiation and 24 cases with low differentiation,19 cases with ?&? stage of TNM and 22 cases ?&?,20 cases with lymph node metastasis and 21 cases with no lymph node metastasis,34 cases with serous infiltration and 7 cases without serosa infiltration.there were no cancer adjacent to carcinoma tissue.All postoperative cases has been confirmed by two pathologists,Using Immunohistochemistry detection and Western blotting technology to test the protein expression NPR-A,P38 MAPK and Cytoskeleton Tubulin-?,and QRT-PCR technique to detect the mRNA expression of NPR-A,P38 MAPK and Cytoskeleton Tubulin-?.We used the Pearson correlation analysis of measurement data and the difference was statistically significant with P<0.05.Results:1 Results of Immunohistochemistry1.1 The expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? protein in gastric normal and cancer tissuesIn gastric cancer tissues expression of P38 MAPK and cytoskeleton Tubulin-? was obviously higher than that of normal gastric tissue(P=0.03,P=0.04),expression of NPR-A in gastric cancer tissue is obviously lower than that in normal gastric tissue(P<0.01).1.2 The relationship between clinic pathological indicators and the protein expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? in gastric cancerThe expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? have no connection with age and sex in gastric cancer(P>0.05).The expression of P38 MAPK and Tubulin-? have no connection with differentiation in gastric cancer(P>0.05).The protein expression of NPR-A of poor differentiated,serous invasion,lymph node metastasis,III and IV stage of TNM in gastric cancer was lower than high-middle differentiation?without serous invasion,without lymph node metastasis,I and II stage of TNM in gastric cancer(P<0.05).The protein expression of P38 MAPK of lymph node metastasis,III and IV stage of TNM in gastric cancer was lower than without lymph node metastasis,I and II stage of TNM in gastric cancer(P<0.05).The protein expression of Cytoskeleton Tubulin-? of serous invasion,lymph node metastasis,III and IV stage of TNM in gastric cancer was higher than without serous invasion,without lymph node metastasis,I and II stage of TNM in gastric cancer(P<0.05).2 The results of Western Blotting2.1 The expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? protein in gastric normal and cancer tissuesIn gastric cancer tissues expression of P38 MAPK and Tubulin-? was obviously higher than that of normal gastric tissue(P<0.01),the expression of NPR-A in gastric cancer tissue is obviously lower than that in normal gastric tissue(P<0.01).2.2 The relationship between clinic pathological indicators and the protein expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? in gastric cancerThe expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? have no connection with age and sex in gastric cancer(P>0.05).The expression of P38 MAPK and Cytoskeleton Tubulin-? have no connection with differentiation in gastric cancer(P>0.05).The protein expression of NPR-A of poor differentiated,serous invasion,lymph node metastasis,III and IV stage of TNM in gastric cancer was lower than high-middle differentiation,without serous invasion,without lymph node metastasis,I and II stage of TNM in gastric cancer(P<0.05).The protein expression of P38 NAPK of serous invasion,lymph node metastasis,III and IV stage of TNM in gastric cancer was lower than without serous invasion,without lymph node metastasis,I and II stage of TNM in gastric cancer(P<0.05).The protein expression of Tubulin-? of serous invasion,lymph node metastasis,III and IV stage of TNM in gastric cancer was higher than without serous invasion,without lymph node metastasis,I and II stage of TNM in gastric cancer(P<0.05).3 Results of QRT-PCR3.1 The expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? mRNA in gastric normal and cancer tissuesIn gastric cancer tissues the mRNA expression of P38 MAPK and Cytoskeleton Tubulin-? was obviously higher than that of normal gastric tissue(P<0.01),expression of NPR-A in gastric cancer tissue is obviously lower than that in normal gastric tissue(P<0.01).3.2 The relationship between clinic pathological indicators and the mRNA expression of NPR-A?P38MAPK and Cytoskeleton Tubulin-? in gastric cancerThe mRNA expression of NPR-A ? P38 MAPK and Cytoskeleton Tubulin-?The expression of NPR-A protein in gastric cancer was positively correlated with the expression of P38MAPK(r = 0.393,P< 0.05),and negatively correlated with the expression of Cytoskeleton Tubulin-?(r =-0.351,P< 0.05),The expression of P38 MAPK protein in gastric cancer was negatively correlated with the expression of Cytoskeleton Tubulin-?(r=-0.047,P<0.01);the expression of NPR-A mRNA in gastric cancer was negatively correlated with the expression of P38 MAPK and Cytoskeleton Tubulin-?(r =-0.581,P< 0.05;r =-0.579,P < 0.05),The expression of P38 MAPK mRNA in gastric cancer was positively correlated with the expression of Cytoskeleton Tubulin-?(r=0.833,P<0.01).Conclusion:1.The protein expression level of NPR-A in gastric cancer tissues was lower than that in normal gastric mucosa tissues,the protein expression levels of P38 MAPK and cytoskeleton tubulin-a in gastric cancer tissues were significantly higher than that in normal gastric mucosa tissues,and NPR-A and P38 MAPK synergistically inhibited the changes of cytoskeleton Tubulin-?.2.In gastric cancer tissues,NPR-A promotes the change of Tubulin-? mRNA by inhibiting the change of P38 MAPK mRNA at the gene level..3.NPR-A can regulate the changes of P38 MAPK signaling pathway and then affect the changes of cytoskeleton Tubulin-?.Therefore,NPR-A can be a potential drug target for targeted therapy of gastric cancer,providing A new idea for the pathogenesis of gastric cancer.
