Objective:To explore the correlation between the expression of TPPP3 in human gastric cancer and the clinical characteristics and prognosis of gastric cancer.Methods:A total of 102 patients who underwent radical gastrectomy(R0 resection)in the Affiliated Zhangjiagang Hospital of Soochow University from July 2014 to January 2017were selected,and their clinical data including age,gender,tumor size,tissue differentiation degree,tumor infiltration depth,regional lymph node metastasis and TNM stage were collected.Any form of preoperative neoadjuvant treatment or translational therapy was not allowed.Using immunohistochemistry,expression of TPPP3 in gastric cancer tissue and adjacent tissue was detected.The associations between TPPP3 expression status and patients'clinicopathological characteristics,and the survival analyses were determined using?~2,survival differences were evaluated with Log-rank test,analysis of variance and Kaplan-Meier method by SPSS 23.0,multivariate analysis were used COX proportional risk regression model.There was a statistical difference when P<0.05.Results:1.The positive expression rates of TPPP3 in gastric cancer tissues adjacent tissues were 77.5%and 34.3%,respectively,which were statistically different(P<0.05).2.The expression status of TPPP3 in gastric cancer tissues was positively correlated with tumor size,regional lymph node metastasis,depth of tumor invasion and TNM staging,with statistically significant differences(P<0.05).However,no significant correlations were detected between TPPP3 expressions and the other clinicopathological characteristics(P>0.05).3.Patients in TPPP3-positive group had significantly lower disease-free survival(DFS)than those in TPPP3-negative group,patients with positive TPPP3 expression in gastric cancer tissues had 1-,2-and 3-year DFS rates of 42.6%,23.3%,and 23.3%after surgery,respectively,while patients with negative expression had 1-,2-and 3-year DFS rates of 79.2%,67.7%,and 67.7%,and the results were statistically different(P<0.05).On the other hand,patients with positive TPPP3 protein expression in gastric cancer tissues had 1-,2-and 3-year overall survival(OS)rates of 78.5%,60%,and 38.5%after surgery,respectively,while patients with negative expression had 1-,2-and 3-year OS rates of78.9%,73.7%,and 67.5%,respectively.The positive expression of TPPP3 in gastric cancer tissues showed a trend of significantly reducing the OS of patients,but the statistical significant difference was not reached(P>0.05).The expression level of TPPP3(P=0.035),depth of tumor invasion(P=0.046)and regional lymph node metastasis(P=0.044)may were independent factors affecting DFS in patients with gastric cancer.Conclusion:TPPP3 is highly expressed in gastric cancer tissues,and its expression level is positively correlated with tumor size,regional lymph node metastasis,depth of tumor invasion and TNM staging.TPPP3 expression may play a role in the occurrence and development of gastric cancer.Highly expressed TPPP3 may increase the risk of postoperative recurrence and be related to poor prognosis of gastric cancer.
