Objective:Based on the small animal MRS(Magnetic Resonance Spectroscopy)technology,the study focus on extracting and comparing the types and contents of important metabolites in the insula area after the treatment of acupuncture for FD(Functional Dyspepsia)at the Shu-Mu points of the stomach,and excavating the key role of insula in the central response of the gastrointestinal regulation upon acupuncture.Methods:1.Thirty 10-day-old male Sprague-Dawley rats were randomly divided into control group,FD model group and FD acupuncture group,with 10 rats in each group,all of which provided breastfeeding.2.The FD model group and the FD acupuncture group were treated with iodoacetamide gavage and tail-clamping method,and irregular feeding method,thus inducing FD models.All the changes were recorded,including weight,general condition and food intake,drinking water.3.During the intervention,the FD acupuncture group was bound daily,and we treat "Zhongwan","Weishu" points with electroacupuncture every day,the frequency was 20~100Hz,2~4V dense waves,20 min/time.After 10 days,we observe the situation of FD rats such as diet by the effect of acupuncture.The FD model group and the control group were only bound without acupuncture.4.After the intervention,rats' superior vena cava blood was taken for detection of gastrin(GAS)in serum by ELISA.The gastric emptying rate was measured by intragastric administration of gastric juice and gastric extraction.5.After anesthesia,the brain tissue of rats was quickly taken out and detected by MRS technique.The TOPSPIN software was matched with the magnetic resonance spectrometer(600 MHz),and the area under the peak was finally determined by the integration method.Then we can compare the absolute concentrations of metabolites such as N-acetyl aspartate(NAA),Aspartic Acid(Asp),Glycine(Gly)and lactic acid(Lac)in the insula(IN).Results:1.After the completion of the models in the two groups,the rats generally showed yellow hairy,loose and niffy stools,weakened activity,getting together and even lying down,and their mood changes from irritatingly high to low.Besides,the body weight,food intake,water intake of the two groups of FD rats were significantly lower than those of the control group(P<0.05).2.After acupuncture intervention,compared with the FD model group,the body weight,food intake,water intake and gastric emptying rate of the FD acupuncture group increased,and the gastrin level decreased significantly.The difference was statistically significant(P<0.05).3.After acupuncture intervention,compared with the FD model group,the concentrations of NAA,Asp,and Gly in the insula area of the FD acupuncture group were significantly lower,and the difference was statistically significant(P<0.05);The difference in the concentration of Lac and other metabolites was not statistically significant(P>0.05).There was no significant difference in the concentration of various metabolites between the FD model group and the control group,which was not statistically significant(P<0.05).Conclusion:1.Acupuncture at Shu-Mu points of the stomach can effectively improve the general condition of FD rats and regulate gastric emptying rate and gastrin level.2.Acupuncture at Shu-Mu points of the stomach can change the content of metabolites NAA,Asp,Gly in the insula significantly,which may be one of the key links in the central response of acupuncture,namely regulating the function of the insula targetedly.
