Objective:In this study,fMRI and functional connection analysis methods were used to explore the influence of Electro-Acupuncture on functional dyspepsia model rats on functional network connection patterns of other relevant brain regions centering on insula functional network,and to further analyze the central mechanism of Stomach Back-Shu and Frontal-Mu Points treatment for FD,in the hope of providing clinical basis for the treatment of FD with Stomach Back-Shu and Frontal-Mu Points.Methods:1.18 7-day old SD lactating mice were randomly divided into control group,FD model group and FD acupuncture group,and 6 lactating mice were included in each group,all of which were males.In the model group and acupuncture group,0.1%iodide acetamide sucrose solution feeding method and guo haijun's tail pinch fighting method were adopted,together with irregular feeding.2.After the model was copied successfully,the FD acupuncture group was captured and fixed on the day after the model was built.Electro-Acupuncture stimulation was performed on Stomach Back-Shu and Frontal-Mu Points.The control group and FD model group were held and fixed with the same method as the acupuncture group,but without acupuncture.3.At the end of acupuncture treatment(/ capture fixation),small animal fMRI scans were performed on each group of rats,and the collected image data were analyzed by functional connectivity analysis method.4.After fMRI scanning,rats in the three groups were given upper chamber venous blood by laparotomy,and then their upper chamber venous blood was centrifuged to obtain serum.The serum gastrin content of rats in each group was determined by ELISA method.Rats were sacrificed after blood collection,and the whole stomach of each group was removed for gastric emptying rate detection.Results:1.Before acupuncture intervention,the weight,food intake and water intake of FD rats in the two groups were significantly lower than those in the control group,with statistically significant differences(P < 0.05).After acupuncture intervention,body weight,food intake and water intake of acupuncture group were significantly improved compared with FD control group,and the difference was statistically significant(P<0.05).2.After the intervention,compared with the FD model group,the gastric emptying rate and gastrin decreased in the FD acupuncture group,and the difference was statistically significant(P<0.05).3.FC analysis results between insula as seed point and whole brain : After intervention,compared with the control group,the functional connection between the model group and the insular lobe was weakened : primary somatosensory cortex,hippocampal CA3,dentate gyrus polymorphic layer,thalamus nucleus,left insular lobe,right insular lobe,external olfactory cortex,parietal cortex,primary visual cortex,and temporal cortex(P < 0.05).After the intervention,compared with the model group,the areas of increased insular functional connectivity in acupuncture group were:primary somatosensory cortex,hippocampal CA3,polymorphous layer of dentate gyrus,caudate putamen nucleus,pons reticular nucleus(P < 0.05).Among them,after the intervention,the acupuncture group compared with the model group had weaker functional connection with the insula:bilateral primary motor cortex,hypothalamus paraventricular nucleus,limbic cortex.
