Objective: Centerd on imbalance in the brain-gut axis mechanism mediated by Ghrelin-centric, to observe electroacupuncture on FD rats generally score, rate of gastric ink residue and small intestine propulsion, ghrelin and Ghrelin receptor expression in gastric antrum, hypothalamus and hippocampus. Investigate the role of Ghrelin in the FD and FD rats mediated by Ghrelin regulation of brain-gut axis imbalances.Methods: Select 80 SPF level SD rats weighting 180 ± 20 g. Before the experiment all rats under general adaptability of squirrel cage after feeding for a week, According to the random number table method,80 rats were randomly divided into normal control group, control group, drug therapy and electro-acupuncture treatment groups, 20 rats in each group. the clip tail building method is adopted to establish the functional dyspepsia model in the rest of three groups of rats other than the normal control group which needs stay away from the building environment to avoid building environmental disturbances. In treatment phase, normal control group and the control group rats daily timed capture, without any other processing; Cisapride tablets in the medication group intragastric administration of dilute solution of cisapride in treatment, daily, continuously for a course of 6 treatments, rest a day after entering the second course, a total of 2 courses; Electric needle treatment group rats daily acupuncture double side "st 36"acupoint(straight thorn 0.3-0.5 inch) and "lr 3"( oblique thorn 0.1-0.2 inch), after twisting needle fast to make sinking astringent sense, Han’s Acupoint Nerve Stimulator was adopted : dilatational wave of constant amplitude, frequency 2/15 Hz, 2m A, the lower extremities of part rats appear slightly trembling. 30 min, daily, continuously for a course of 6 treatments, rest a day after entering the second course, a total of 2 courses. Observing the effection of electric acupuncture in functional dyspepsia rats on generally score、daily body weight gain、eating and water volume 、Gastric residual volume、intestinal ink advance rate and related indicators. Pathological changes in the gastric antrum smooth muscle tissue was observed using a microscope; plasma concentrations of Ghrelin values was detected by enzyme-linked immunosorbent assay; The expression of Ghrelin protein in stomach、hypothalamus and hippocampus of each group rats were detected by immunohistochmistry staining;Real-time fluorescent quantitative PCR method was used observing the expression of GHS-R m RNA in stomach、hypothalamus and hippocampus of each group ratsResults:(1)Compared with normal control group, FD rats generally score in the modeling phase, daily increased body weight and daily food intake, daily water intake were significantly lower(P<0.01); During the treatment phase, with electro-acupuncture and drug treatments, electro-acupuncture treatment group and drug treatment group rats generally scores、daily increased body weight 、daily food intake、 daily water intake were increased, and a significant difference compared with the Group modeling phase(P<0.05,P<0.01); Moreover, the electro-acupuncture treatment group rats grew further increase in weight compared with the drug therapy group(P<0.01).(2) Gastrointestinal tissue on morphology found no organic changes in each group rats, the colours of gastric mucosa in model grouponly being white observed by the naked eye. Other three groups of rats stomach mucosal found no ulcer、hemorrhage or inflammation, under the light microscope(x400) we found that only model group has a few neutrophils in gastric tissue.(3) compared to normal control group, FD rats gastric residual volume increased(P<0.05), intestinal ink advance rate decreased(P<0.01); Compared with control group, electro-acupuncture treatment group and drug treatment group rats gastric residual volume were significantly reduced(P<0.01), intestinal ink advance rate significantly incerased(P<0.01);and it has no significant difference between electro-acupuncture treatment group and drug treatment group.(4) Compared with normal control group, FD model rats of ghrelin concentrations in peripheral blood plasma、gastric mucosa、hypothalamus 、hippocampus were decreased(p <0.05), electroacupuncture can raise Ghrelin concentrations in peripheral plasma 、stomach tissues、hypothalamus and hippocampus in FD rats(P<0.05, and P<0.01), however, the drug of cisapride solution only raise ghrelin concentrations in gastric tissues(P<0.05),but cannot raise Ghrelin levels in the peripheral blood plasma、 hypothalamus and hippocampus(P>0.05).(5)Compared with the control group, GHS-R m RNA expression in gastric tissue and hypothalamus decreased in FD rats(P<0.05); Compared with the control group, the expression of GHS-R m RNA in gastric tissue、hypothalamus and hippocampus increased significantly in electro-acupuncture treatment group(P<0.05); however, the drug treatment group only raise GHS-R m RNA expression in stomach and hypothalamus, but not obvious in hippocampus.Conclusion:(1) Clip tail method copy FD rats meets diagnostic criteria for FD; FD models rats generally score、gastric residual volume increased, intestinal ink advance rate declined, ghrelin concentrations in the peripheral plasma 、gastric mucosa、 hypothalamus 、 hippocampus and GHS-Rm RNA expression in gastric tissues and hypothalamus all decreased;(2) Electro-acupuncture and cisapride treatments can be effective in improving general conditions、increasing daily weight gain、 daily eating and drinking volume;(3)the function of gastric emptying and small intestine motility declined in FD model rats, both electro-acupuncture and cisapride can promote gastric emptying and small intestine motility function, No significant differences between the two groups;(4) the expression of ghrelin in peripheral blood plasma and gastric mucosa 、 hippocampus and hypothalamus decreased in FD model rats, electroacupuncture can promote increasing ghrelin concentrations in peripheral plasma 、stomach tissues、hypothalamus and hippocampus in FD rats, however, the drug of cisapride solution only promote the expression of ghrelin in gastric tissues, but have no effect on ghrelin levels in the peripheral blood plasma、hypothalamus and hippocampus.(5) Electroacupuncture treatment can promote the expression of GHS-R m RNA in gastric tissue 、 hypothalamus and hippocampus,however, the drug treatment group only raise GHS-R m RNA expression in stomach 、hypothalamus, the effect of GHS-R m RNA expression on hippocampus is not obvious.Therefore, we believe that: Drugs and electro-acupuncture can improve the general situation 、 food intake 、 the expression of Ghrelin and GHS-Rm RNA in gastric tissues, promote increasing expression of GHS-Rm RNA in hypothalamus and hippocampus; Compared with the drug treatment group, electro-acupuncture can increase plasma Ghrelin levels, promoting the expression of Ghrelin in hypothalamus, increasing GHS-Rm RNA expression in gastric and hippocampus. Study shows that, compared with drugs, electric acupuncture has advantages of multi-aspect 、bi-directional adjustment and so on, electroacupuncture can better regulate brain-gut axis with multi-targetes and to better improve the FD symptoms.
