Objective:1.This study was to explore the gene transcription profiles of left and right colon cancer,providing molecular genetic basis for the difference in biological characteristics of left and right colon cancer.2.This study was to compare the commonmolecular pathological markers in left and right colon cancer,verifying the difference in gene expression between left and right colon cancer,for clarifying its clinical significance.Methods:1.Three left and right colon cancer tissues and adjacent tissues were selected,and a total of 12 samples were divided into 4 groups,including left colon cancer tissue and right colon cancer tissue,left colon cancer adjacent tissue and right colon cancer tissue,left colon cancer tissue and left colon cancer adjacent tissue,right colon cancer tissue and right colon cancer adjacent tissue.The extracted total RNA samples were subjected to quality inspection and quantification,and be used to construct library and sequence.Then cluster the differentially expressed genes,GO significantly enriched and Pathway significantly enriched analysis.2.The clinical data of 73 patients with left colon cancer and 67 patients with right colon cancer were retrospectively analyzed.The differences in biological characteristics between left and right colon cancer were compared.The differentially expressed genes of left and right colon cancer were verified by clinically used immunohistochemical molecular markers,exploring its clinical significance.Results:1.There are many differences of gene transcription profiles in left and right colon cancer,involving multiple signaling pathways,including oxidative phosphorylation,citric acid cycle,cytokines and cytokine receptors,MAPK signaling pathway,PPAR signaling pathway,P53 signaling pathway,APC signaling pathway and so on.Compared with the right colon cancer tissues,the upregulated genes in left colon cancer tissues are TP53I11,MSI1,ANAPC15,BCLAF3,COX7 B,etc.And the down-regulated genes are SNAPC2,MSLN,PRSS3,TGFB1,PLK3,VEGFC,VGF,BCL2A1,etc.2.The results of clinical data analysis showed that there were gender differences between left and right colon cancer,and the first clinical symptoms were significantly different.The symptoms of blood,intestinal obstruction and change of stool habits in left colon cancer were more common than those in right colon cancer,while the symptoms of abdominal pain,emaciation and anemia were less common than those in right colon cancer(P < 0.05).Pathological type,mucus composition,maximum diameter and gross morphology of cancer patients were significantly different.The left colon cancer was mainly moderately differentiated adenocarcinoma,while the right colon cancer was mainly moderately and poorly differentiated adenocarcinoma.The proportion of mucus type and maximum diameter of cancer was higher than that of left colon cancer,and the proportion of ulcerative type was lower than that of left colon cancer(P < 0.05).There was no significant difference in staging,nerve invasion,intravascular tumor thrombus and length of history between the two groups(P > 0.05).There was a significant difference in the rate of poor prognosis between the two groups(P < 0.05).Among the common molecular pathological markers,the positive expression of MLH1,MSH2,P53 in left and right colon cancer was significantly different(P < 0.05),while the positive expression of COX-2,VEGF,Her-2,EGFR,Ki-67 had no statistical significance(P > 0.05).There is a phenomenon that both groups had high expression of COX-2 and EGFR.Conclusions:1.There are significant differences of gene transcription profiles between left and right colon cancer,which is the molecular genetic basis of biological characteristics in left and right colon cancer.It lays an experimental foundation and provides a theoretical basis for further exploring the pathogenesis of left and right colon cancer and realizing individualized targeted therapy.2.There are differences in gender,clinical symptoms and pathological types between left and right colon cancer.Both of them highly express COX-2 and EGFR.The expression of MLH1,MSH2 and P53 was significantly higher than those in right colon cancer.These common molecular pathological markers are of great significance in guiding precise individualized treatment of clinical colon cancer.
