Objective:This study conducted a meta-analysis of the published studies on the association between the polymorphism of rs1256049 locus of estrogen receptor ? gene and the risk of prostate cancer at home and abroad,so as to obtain more accurate and objective conclusions,provide more reliable basis for basic research and clinical treatment,and achieve the purpose of predicting risks in advance,assisting drug development and preventing diseases.Methods:All relevant literatures from the self-established database to February 2019 were retrieved by computer in PubMed,Embase,Cochrane library,cnki and wanfang databases.The retrieved literatures were carefully screened according to the inclusion criteria and exclusion criteria,and the quality control was carried out by means of double review.Data were extracted from the included studies,and the Review Manager 5.3 was applied to analyze the data.Heterogeneity tests were performed on each study.According to the heterogeneity test results,the corresponding effect model was selected to calculate the combined advantage ratio(OR)and its 95% confidence interval(Cl).Subgroup analysis was conducted according to the population ethnic type,and sensitivity analysis was conducted by using the transformation effect model and excluding the most heavily weighted literatures one by one.Funnel plot was used to analyze publication bias.Results:A total of 42 related literatures were retrieved in this study,and 6 literatures and 8studies were finally included after screening,all of which were case-control studies.Among them,four studies were conducted in Asian populations,three in Caucasian populations and one in African populations.There were 7796 cases in the prostate cancer group and 8927 cases in the control group.rs1256049 polymorphism of estrogen receptor ? gene was associated with prostate cancer risk in the general population(AA vs GG:OR=1.40,95% Cl=1.02-1.91;AA vs GG+GA:OR=1.50,95% Cl=1.10-2.04).Subgroup analysis based on ethnicity showed that ER rs1256049 polymorphism was significantly correlated with prostate cancer risk in Caucasian population(A vs G:OR=1.19,95% Cl=1.06-1.35;AA vs GG: OR=2.18,95% Cl=1.17-4.07;AA vs GG+GA: OR=2.20,95% Cl=1.18-4.10;GA+AA vs GG:OR=1.17,95% Cl=1.03-1.33).Asian population and African population showed no correlation,which may be related to the small number of studies.Conclusion:The polymorphism of rs1256049 of estrogen receptor ? gene is correlated with prostate cancer susceptibility,especially with prostate cancer susceptibility in Caucasian population,which can promote the risk of prostate cancer in Caucasian population.The association between prostate cancer susceptibility in Asian and African populations needs further study.
