Objective:To systematically evaluate the relationship between the polymorphism of cyclooxygenase-2(COX-2)-8473T>C(rs5275)gene and susceptibility of prostate cancer.Methods:The three common English databases of Pubmed,Embase,Cochrane Library and three common Chinese databases of Chinese biomedical literature database,CNKI and Wanfang were searched to collect case-control studies on COX-2-8473T>C(rs5275)gene polymorphism and prostate cancer susceptibility.The retrieval time was limited to the database establishment to February 11,2020.The literatures were screened strictly according to inclusion and exclusion criteria,and the literatures were screened independently by 2 evaluators to extract relevant useful data for the included studies.Data were processed by Rev Man5.3 software,heterogeneity test was conducted for the included studies,accurate effect model was selected,and 95% confidence interval(CI)and odds ratio(OR)were calculated.Results:In the end,8 articles(9 studies)were included in this Meta-analysis,all of which were case-control studies.The Caucasian,Asian,and African Populations include 5,3,and 1 studies,respectively.The number of the prostate cancer group and the control group were 12967 and 13516,respectively.COX-2-8473T>C(rs5275)gene polymorphism was correlated with the susceptibility of prostate cancer in the general population(CC VS TT: OR=1.30,95%Cl=1.02-1.67,p=0.04).Subgroup analysis based on ethnic type showed that COX-2-8473T>C(rs5275)gene polymorphism was correlated with prostate cancer susceptibility in Asian population(C VS T: OR = 1.24,95% Cl = 1.05-1.46,p = 0.01).CC VS TT: OR = 1.76,95% CI = 1.18-2.61,p = 0.005;CC VS TC + TT: OR = 1.65,95% Cl = 1.15-2.37,p = 0.007),and had no correlation with Caucasian and African populations.Conclusion:The CC genotype of COX-2-8473T>C(rs5275)in the general population can increase the risk of prostate cancer,the Asian population with the C allele at the COX-2-8473T>C(rs5275)has an increased risk of prostate cancer.However,there is no correlation between COX-2-8473T>C(rs5275)gene polymorphism and prostate cancer risk in Caucasian and African populations.
