Purposes: MiRNA(microRNA),as a family of non-coding small RNA 19-25 nt in length,have been proved to play abroad roles in the development of organism involving many aspects of physiological and pathological processes such as organ formation,cell proliferation,tumor and so on.Periodontitis and peri-implantitis are the primary cause of the looseness and loss of teeth and implants.Therefore,in this study,we attempt to explore the expression profiles of miRNAs in the two inflammation tissues by establishing canine models,as the groundwork of the bone tissue engineering complex for repairing defect of inflammatory.Methods: Induce periodontitis and peri-implantitis six adult labrador dogs with silk-wire twisted ligature forced to the gingival margin of teeth and implants.Then extract the healthy gingival tissue,periodontitis tissue and peri-implantitis tissue for miRNA sequencing and compare their differences.Screen the differently expressed miRNAs for real-time PCR in order to prove the reliability of miRNA sequencing.Results: The results show that there are significant differences of miRNA expression among the three kinds of tissues.Screen the cfa-let-7g、cfa-miR-27a、cfa-miR-29 a and cfa-miR-142 for real-time PCR and trends of cfa-let-7g、cfa-miR-27 a and cfa-miR-29 a agree with the miRNA sequencing and the different expressions of cfa-miR-27 a were statistically significant(p<0.05).Conclusion: We demonstrated that many miRNAs expressed differently among the healthy gingival tissues,periodontitis tissues and peri-implantitis tissues in this study.cfa-let-7g、cfa-miR-27a、cfa-miR-29 a and cfa-miR-142 may closely relate to the development of periodontitis and peri-implantitis.Combining to the results of real-time PCR,the expression of cfa-miR-27 a in the inflamed tissue was obviously lower compared to the healthy tissues,indicating that cfa-miR-27 a may involve in downstream biological processes through its target genes.
