| Objective: To observe the expression of Notch3 mRNA and Jagged2 mRNA in the lungs of chronically asthmatic rats and to elucidate the effect of Pingchuan Granules on airway remodeling in rats with chronic bronchial asthma.Methods: Sixty female Wistar rats were randomly divided into the blank group,the model group,the dexamethasone group and the Pingchuan Granules group.Except for the blank group,other three groups were sensitized and stimulated with the solution of chicken ovalbumin and aluminum hydroxide to establish the rat model of chronic asthma.Five days after the last sensitization,dexamethasone(1mg/kg)was given to the dexamethasone group,Pingchuan Granules(5ml/kg)was given to rats in the Pingchuan Granules group,normal saline(10ml/kg)was given to the blank group and the model group.The rats were killed 24 hours after the last challenge,the changes of airway remodeling in lung tissue of asthma rats would be observed under light microscope.The expression of Notch3,Jagged2 mRNA in lung tissue would be detected by Real-Time PCR.Results:1.The general state of the rats in each group: After intragastrically administration,the respiration,hair,activity,reaction,diet and body weight of the rats in the blank group were normal.In the model group,the respiratory rate was accelerated,cough and wheezing symptoms appeared,hair dropped,activity and reaction decreased,diet decreased and weight gain slowed down;The state of rats in dexamethasone group was improved compared with that in model group;At initial state of using Pingchuan Granules,the symptoms were not significantly improved compared with the model group,but in the later stage,the symptoms of the Pingchuan Granules group were significantly better than those of the dexamethasone group.2.Pathological changes of lung tissue in each group under lightmicroscope: There were no obvious pathological changes of airway remodeling in the blank group;Compared with the blank group,there were many pathological features of airway remodeling such as obvious inflammatory cell infiltration around the bronchus and blood vessels,disordered arrangement of bronchial epithelial cells,necrosis and shedding,edema of airway mucosa,proliferation,hypertrophy of bronchial smooth muscle,dilatation and increase number of blood vessels in the lung tissue of rats,proliferation and thickness increase of vascular wall and changes of alveolar pulmonisin the model group;The pathological changes of airway remodeling in Pingchuan Granules group and the dexamethasone group were significantly less than those in model group,especially in Pingchuan Granules group.3.Pingchuan Granules could increase the expression of Notch3 mRNA in the lungs of rats with chronic asthmatic bronchial asthma,which is significantly different from that of the model group.There is no difference with the dexamethasone group.4.Pingchuan Granules could reduce the expression of Jagged2 mRNA in lung tissue of rats with chronic asthma in bronchial asthma,and there is significant difference compared with the model group,and there is no difference with dexamethasone group.Conclusions:1.Pingchuan Granules could increase the expression of Notch3 mRNA in lung tissue of rat models of chronic persistent asthma.2.Pingchuan Granules could reduce the expression of Jagged2 mRNA in lung tissue of rat models of chronic persistent asthma.3.Pingchuan Granules could improve airway remodeling in rat models of chronic persistent asthma. |
