Objective:To observe the effects of semen arecae.charred semen arecae.semen arecae-removal.charred semen arecae-removal on gastrointestinal motility.We test GIP,GLP-1,gastric and bile secretion in rats.The mechanism of“burnt odour arouse the spleen”provides a reference for the clinical application of Chinese medicine.Method:Sixty SD rats were randomly divided into 6 groups.10 rats were divided into each group,which includes half males and half females.There are some groups,such as Control Group,semen arecaecharred semen arecae.,semen arecae-removal.,charred semen arecae-removal,positive drug group?positive group was administered mosapride?.After 7 days of adaptive feeding,each group of Semen Arecae was intragastrically administered at 3 g/kg?equivalent to 10 times the clinical dose?.The control group was intragastrically administered with normal saline,and the positive group was administered with mosapride at a dose of 2.5 mg/kg.Seven days after the administration,blood was taken from the abdominal aorta,the blood was centrifuged to obtain serum,and the contents of GIP and GLP-1 were measured by ELISA method,followed by gastric emptying and intestinal propulsion experiments.In addition,rat gastric juice and bile were collected,and the amount of gastric secretion,pH,pepsin activity,bile amount,cholesterol,bile acid,and bilirubin were measured.Result:1.Effect of gastric emptying and intestinal propulsion in rats of the groups of semen arecaeCompared with the control group,the gastric emptying ability of charred semen arecae,charred semen arecae-removal and positive drug group showed a significant increase(??P<0.01).The gastric emptying ability of charred semen arecae group was significantly higher than semen arecae group(??P<0.01).The gastric emptying ability of charred semen arecae-removal was significantly higher than semen arecae-removal?**P<0.01?,Compared with charred semen arecae and charred semen arecae-removal were enhanced in the gastric emptying ability.But the diference was not significant.Compared with the control group,semen arecae,charred semen arecae,semen arecae-removal.charred semen arecae-removal.positive drug group showed a significantly increase in intestinal propulsion(??P<0.01).The intestinal propulsion capacity of the charred semen arecae group was significantly higher than that semen arecae.(??P<0.01).charred semen arecae-removal was significantly higher than semen arecae-removal?**P<0.01?,Compared with charred semen arecae and charred semen arecae-removal were enhanced in intestinal propulsion.But the diference was not significant.2.Effect of gastric secretion,pH and pepsin activity in rats of the groups of semen arecaeComparing the control group,there was no difference in gastric secretion and pH between semen arecae,charred semen arecae,semen arecae-removal.charred semen arecae-removal.positive drug group?P>0.05?.The difference in pepsin activity was extremely significant and were significantly increased compared with the control group?**P<0.01?.The pepsin activity of charred semen arecae was significantly higher than semen arecae(??P<0.01).The pepsin activity of charred semen arecae-removal was significantly higher than semen arecae-removal??P<0.01).The pepsin activity of charred semen arecae and charred semen arecae-removal were enhanced.But the difference was not significant.3.Effect of bile,cholesterol,bile acid and bilirubin in rats of the groups of semen arecaeComparing the control group,semen arecae,charred semen arecae,semen arecae-removal.charred semen arecae-removal.positive drug group,the bile acid was not different between the groups?P>0.05?,But the difference in bile secretion,cholesterol,and bilirubin was significant.The bile secretion of charred semen arecae,charred semen arecae-removal and positive drug group were significantly higher than control group?*P<0.05?.The bile secretion in the charred semen arecae was higher than semen arecae??P<0.05?.The bile secretion in the charred semen arecae-removal was higher than semen arecae-removal??P<0.05?,Compare with charred semen arecae and charred semen arecae-removal were increased.But the difference was not significant.The bilirubin of semen arecae,charred semen arecae and charred semen arecae-removal were significant higher than control group?**P<0.01?.The cholesterol of charred semen arecae,charred semen arecae-removal and positive drug group were significant higher than control group?**P<0.01?.The cholesterol and bilirubin of charred semen arecae was significantly higher than semen arecae(??P<0.01).The cholesterol and bilirubin of charred semen arecae-removal was significant higher than semen arecae-removal(??P<0.01),.The cholesterol and bilirubin of charred semen arecae and charred semen arecae-removal were increased.But the difference was not significant.4.Effect on GIP and GLP-1 levels in rats of the groups of semen arecaeCompared with the control group,the GIP content in rat serum of semen arecae,charred semen arecae,semen arecae-removal.charred semen arecae-removal.positive drug group were extremely significant increased?**P<0.01?.The GIP content in rat serum of the charred semen arecae was significantly higher than semen arecae,(??P<0.01),The difference between semen arecae-removal and charred semen arecae-removal was not significant,in the GIP content in rat serum.The difference in charred semen arecae and charred semen arecae-removal are also not significant enough.Compared with the control group,the GLP-1 content in rat serum of charred semen arecae,semen arecae-removal.charred semen arecae-removal.positive drug group were extremely significant increased?**P<0.01?.The GLP-1 content in rat serum of semen arecae was higher than control group?*P<0.05?.The GLP-1 level in the rat serum of the charred semen arecae was significantly higher than semen arecae(??P<0.01).The charred semen arecae-removal was significantly higher than the semen arecae-removal(??P<0.01).There was no significant difference between the group of charred semen arecae and the charred semen arecae-removal.Conclusion:The ability of gastrointestinal exercise,pepsin activity,bile secretion,bile cholesterol and bilirubin content,and GLP-1 levels in the charred semen arecae group and the charred semen arecae-removal were significantly higher than semen arecae and semen arecae-removal.The difference between the charred semen arecae and charred semen arecae-removal was not significant.After the semen arecae is fried,the"burnt odour arouse the spleen"and the main component of semen areca are not related to alkaloids.It may be related to the Maillard reaction after the semen arecae is fried to promote the secretion of GLP-1,increase the activity of pepsin and bile.The amount of secretion of bilirubin and cholesterol promote gastrointestinal motility.
