Chinese medicine is the essence of the ancient wisdom of the ancient,its historical status and social value is irreplaceable.Traditional Chinese medicine is an important part of Chinese medicine,and its quality is an important guarantee of stable development of Chinese medicine.In recent years,traditional Chinese medicine is widely used in various industries because of its unique medicinal value,which lead to the increasing demand.To meet the needs of the consumers,cultivated products had been become the main source of traditional Chinese medicine.However,there was no strict,unified planting,processing and other norms that make some problems for the quality of traditional Chinese medicine.In recent years,a variety of exogenous pollutants in different Chinese medicine were constantly detected,which caused a serious problem for the quality,effectiveness and safety of traditional Chinese medicine.Mycotoxins are one of the exogenous harmful substances,and they can accumulate in the body so that to cause cancer or damage liver and kidney function,seriously threat to human health and life safety.Traditional Chinese medicine is easy to mold and produce mycotoxins that effected by herbs own external factors and external environment in the storage process.Therefore,it is a great significance that the traditional Chinese medicine stored in the appropriate environment and make good maintenance for preventing traditional Chinese medicine mold,quality changes,to ensure the quality of traditional Chinese medicine,effectiveness and safety.In this study,Mentha haplocalycix herba,citri reticulatae pericarpium and Semen Arecae were used as the research medicines.The characteristics of the traits,water content,the contents of active ingredients and the accumulation of mycotoxins were selected as evaluation indexes,stored mendicines under different storage environments(high temperature and humidity,low temperature and humidity,room temperature and humidity)and packaging materials(self-styled bags,kraft paper,woven bags)for 12 months,then were examined the changes of the quality of medicinal herbs before and after storage and mildew,so as to find that suitable storage environment and packaging materials for traditional Chinese medicine storage,and also,to provide a theoretical basis for ensuring the quality of traditional Chinese medicine and establishing storage norms.The main contents of this thesis are as follows:1.Effects of different storage conditions on the quality of Mentha haplocalycis herba.The UPLC-PDA method was established to simultaneous determination of eight active ingredients(caffeic acid,hesperidin,rosmarinic acid,diosimin,diosmetin-7-O-glucopyranoside,didymin and buddleoside).The traditional QuECHERS method was optimized and the UFLC-MS/MS method was used to detect 15 mycotoxins(aflatoxin(AFB1),B2(AFB2),G1(AFG1),G2(AFG2),ochratoxin A(OTA),HT-2 toxin(HT-2),T-2 toxin(T-2),fumonisin B1(FB1),fumonisin B2(FB2),citrinin(CIT),zearalanone(ZAN),alternariol(AOH),alternariol monomethyl ether(AME)and tentoxin(TEN),zearalenone(ZEA))in Mentha haplocalycis herba.The effects of different storage conditions on the quality of Mentha haplocalycis herba were investigated by the changes of traits,water content and the contents of active ingredient and mycotoxins.The results showed that Mentha haplocalycis herba was lighter in color and had no mildew and moth-like phenomenon after 12 months storage in different storage environments and packaging materials.The content of water content and active ingredient changed with the storage time.AOH,AME,TEN and ZEA were detected in all samples,at the same time,AFB1,AFB2,AFG2,FB1 and FB2 was also found,but the content was low under low temperature and temperature environment and self-contained bag,therefore it was suggested that the preferred packaging materials for Mentha haplocalycis herba storage in self-styled bags under low temperature and humidity.2.Effects of different storage conditions on the quality of citri reticulatae pericarpium.The characteristics of traits,water content,the contents of active ingredients(naringin,hesperidin,sinensetin,naringin,tangeretin)and aflatoxins were used as observation indexes.The UPLC-PDA method and UFLC-MS/MS method were used to study the effects of different storage conditions on the quality of citri reticulatae pericarpium.The results showed that color of citri reticulatae pericarpium was deepened,no mildew phenomenon,the sample storage in kraft paper and woven bags had varying degrees of mothy phenomenon after 12 months storage.Water content increased more than pharmacopoeia standards.The contents of active ingredients were decreasing,especially for the content of hesperidin did not meet the pharmacopoeia standard after storage.All samples were not detected aflatoxins.According to the changes for the characteristics of traits,water content,contents of active ingredient,self-styled bags and low temperature and humidity environment is more suitable for storage of citri reticulatae pericarpium.3.Effects of different storage conditions on the quality of Semen Arecae.By using the UPLC-PDA method to detect the contents of arecoline,guvacine and arecaidine in Semen Arecae,the contents of total phenolics,total flavonoids and polysaccharides were determined by UV spectrophotometry.11 mycotoxins in Semen Arecae were detected by UFLC-MS/MS method.And observed the characteristics,the water content to investigate the effects of different storage environment and packaging materials on the quality of Semen Arecae.The results showed that after 1 year of storage,the color of Semen Arecae was deepened,no mildew and moth,and the water content was increased.The contents of active ingredients were declined,but the decline for the contents of active ingredients of Semen Arecae in low temperature and humidity environment and self-styled bags was little;11 mycotoxins were all not detected.It is recommended that the self-contained bag and low temperature and humidity environment is benefit for the storage of Semen Arecae.
