Objective:This experiment through the observation of coke Betel nut extract, knocking out the focal Betel nut alkaloids extract, and adding different receptor and adding coke Betel nut extract on gastrointestinal smooth muscle contraction body from rats, to explore the mechanism of coke Betel nut on gastrointestinal movement possible.Method:Preparation of isolated rat gastrointestinal smooth muscle specimens, the Betel nut extract for the control group, the organization was measured on isolated gastrointestinal smooth muscle tension before and after dosing system, to observe the effects of different concentrations of coke Betel nut extract, knockout alkaloids after focal Betel nut effects of aqueous extract liquid on gastrointestinal smooth muscle muscle strips, and compared in the accession to the M, N, alpha, beta, receptor agonist or antagonist and Ca2+antagonist after focal Betel nut extract effect change.Result:l.Coke Betel nut effect of water extract on the contraction of gastrointestinal smooth muscle of rats in vitroAnd the concentration of water extract of raw Betel nut compared. Betel nut coke water extract on gastrointestinal smooth muscle tension of isolated rat increased significantly, and the concentration of Betel nut extract were compared (P<0.05); Betel nut coke water extract on isolated gastrointestinal smooth muscle tension shows a dose-dependent among them, the dose of20μLand80μL, on gastric and intestinal increased more obviously the amplitude, frequency speed (P<0.01).2.Knockout effects of alkaloids from the focal Betel nut extract on the contraction of gastrointestinal smooth muscle of rats in vitroWith the concentration Betel nut coke water extract were compared, knockout alkaloids Betel nut coke water extract on gastrointestinal smooth muscle contraction were significantly decreased, the amplitude becomes small, slow frequency (P<0.05); and the concentration Betel nut coke water extract group, there were significant differences in80μL, tension decreased. More obvious (P<0.01).3.Effects of M receptor agonists and antagonists focusing Betel nut water extract induced contraction of isolated gastrointestinal smooth muscle bodyCompared with the students Betel nut extract group, atropine sulfate can inhibit the students Betel nut extract strongly spasm on gastrointestinal smooth muscle, reduce the amplitude and frequency of motility (P<0.01), acetylcholine chloride can obviously promote the students Betel nut extract on intestinal smooth muscle contraction tension bar (P<0.01); and the focal Betel nut extract group, atropine sulfate induced focal Betel nut water extract on gastrointestinal smooth muscle was significantly inhibited (P<0.01), contractile effect of acetylcholine chloride could induce focal Betel nut Decoction on intestinal smooth muscle (P<0.01). And Betel nut extract group, acetylcholine chloride induced focal Betel nut extract has a stronger effect on promoting on isolated intestinal smooth muscle stripes, the effect of the stomach does not change obviously.4.Effects of N receptor agonist focusing Betel nut water extract induced contraction of isolated gastrointestinal smooth muscle bodyCompared with the students Betel nut extract group, nicotine can significantly promote Betel nut extract liquid on intestinal smooth muscle spasm strips, contraction decreased significantly (P<0.01); and the focal Betel nut extract group, nicotine could reduce the coke Betel nut water extract on gastrointestinal smooth muscle tension, but without statistical significance (P>0.05). Born with betel nut water extraction liquid ratio, nicotine induced focal betelnut water extraction liquid of in vitro with weak gastrointestinal smooth muscle spasm.5.Alpha receptor agonists and antagonists focusing effect of extraction from contraction in gastrointestinal smooth muscle induced Lang DecoctionAnd Betel nut extract group, phentolamine mesylate can obviously promote the students Betel nut extract liquid on gastric smooth muscle contraction tension of strip (P<0.01), nor adrenaline bitartrate can make intestinal a smooth muscle relaxation, tension decreased, contraction frequency and amplitude decreased (P<0.01); and the focal Betel nut extract liquid group, phentolamine mesylate also can promote the Betel nut coke water shrinkage tension liquid on gastric smooth muscle stripes (P<0.05), noradrenaline bitartrate induced focal Betel nut extract liquid on intestinal smooth muscle tension bar decreased significantly (P<0.01). Compared with raw betelnut water extraction liquid group, phentolamine mesylate and tartaric acid induced focal norepinephrine betelnut water extract on gastrointestinal smooth muscle tension variation is not obvious. 6.Beta agonists and antagonists focusing effect of extraction from contraction in gastrointestinal smooth muscle induced Lang DecoctionAnd Betel nut extract group, propranolol hydrochloride can obviously promote the students Betel nut water shrinkage effect liquid on gastric strips, tension increased significantly (P<0.01), isoprenaline hydrochloride can be blocked by students Betel nut extract liquid on intestinal smooth muscle tension bar (P<0.01); group compared with the Jiao Binlang water extraction, propranolol hydrochloride can be induced by focal Betel nut extract increased tension on gastrointestinal smooth muscle (P<0.05), isoprenaline hydrochloride induced focal Betel nut extract on gastrointestinal smooth muscle tension decreased significantly (P<0.01). Compared with raw betelnut water extraction liquid group, propranolol hydrochloride induced focal betelnut water extraction liquid of intestinal article effect more apparent.7.Effect of Ca2+antagonist focusing Betel nut water extract induced contraction of isolated gastrointestinal smooth muscle bodyAnd Betel nut extract group, Veerappa Mikko hydrochloride significantly induced by students Betel nut water extract on intestinal smooth muscle stripes tension decreased significantly (P<0.01); and the focal Betel nut extract group, Vera Pammy hydrochloride can obviously block the contractile tension on the intestinal smooth muscle stripes (P<0.01); Um Vera Pammy salt water extract and induce Betel nut Betel nut coke water extract on gastric smooth muscle tension was decreased the role, but had no statistical significance (P>0.05). Compared with raw betelnut water extraction liquid group, verapamil hydrochloride induced focal betelnut water extraction liquid of gastrointestinal smooth muscle in vitro of change is not obvious.Conclusion:1. Coke Betel nut extract can promote rat gastrointestinal smooth muscle tension, and showed a dose dependent, the stomach of the role more obvious; by knocking out the contrast before and after that alkaloids, alkaloids are the main material basis for promoting gastrointestinal motility.2. Through the study of coke Betel nut water extract on gastrointestinal smooth muscle function from the rats, and the mechanism can be summarized as follows:(1) By comparing the added acetylcholine chloride and atropine sulfate after analysis, Ach agonists on M receptor, resulting in extracellular Ca2+influx, increased Ca2+concentration in cytosol, muscle membrane depolarization, and gastrointestinal smooth muscle contraction;(2) By adding a comparative analysis before and after nicotine, nicotine has inhibitory effect on gastrointestinal smooth muscle may be weakened, activation of N receptor induced release of NO causes muscle contraction activity;(3) by the addition of phentolamine mesylate and noradrenaline bitartrate were compared before and after, noradrenaline bitartrate activated receptor alpha, promote K+outflow, the hyperpolarization produced muscle membrane, resulting in relaxation of gastrointestinal smooth muscle;(4) By adding contrast before and after propranolol hydrochloride and isoprenaline hydrochloride, may be relaxed sympathetic release of norepinephrine and beta two receptor binding to smooth muscle;(5) By adding hydrochloric acid before and after contrast Vera Pammy, may be blocked by Ca2+, inhibition of smooth muscle contraction.3. Focal betelnut water extract on gastrointestinal smooth muscle the promoting effect is much better than that of raw betelnut water extraction liquid, its mechanism may be related to M, N, beta receptor pathway.
