Objective:This preliminary study was based on the good clinical effect of acupoint catgut implantation and the regulation of neurogenic inflammation.The influence of Acupoint catgut embedding on the sensory neuropeptide substance P and MC,and the influence of Acupoint catgut embedding on the concentration tolerance and the time tolerance of OVA allergens were taken as the research direction to further explore the mechanism of Acupoint catgut embedding on MPI/AR.It provides a more reliable theoretical basis for Acupoint catgut embedding in the clinical treatment of MPI/AR and AR.Methods:1.The SD rats AR model and the SD rats MPI/AR model were established with OVA.The SD rats were randomly divided into the blank control group,the AR model group,the MPI/AR model group,acupoint catgut embedding group,the capsaicin group,six for each group.Acupoint catgut embedding group and the capsaicin group were based on the MPI/AR model,respectively used the nasal intervention of the puncture buried line and the capsaicin.After 2 weeks of intervention,observed their Behavioral,collected their serum(Elisa test)and took out their nasal mucosa tissue(He staining,Toluamine blue dye,Immunohirochemical detection,Western blot test).2.The SD rats MPI/AR model was established with OVA.The SD rats were randomly divided into the MPI/AR model group,acupoint catgut embedding group and the fake buried line group,sixteen for each group.Acupoint catgut embedding group and the fake buried line group were based on the MPI/AR model,respectively used the nasal intervention of the puncture buried line and the fake buried line.After 2 weeks of treatment,observed and counted the After 2 weeks of treatment.The three sets of models were divided into A group and B group.The various groups of A were conducted to perform the test of the concentration tolerance of OVA and the various groups of B were conducted to perform the test of the time tolerance of OVA.Results:1.The effect of acupoint catgut embedding on the sensory neuropeptide SP and MC.(1)Among all the detection and statistical results of the various SD rats were statistically significant(P<0.001).The AR model group was significantly higher than the other groups,and the difference was statistically significant(P<0.05).(2)The statistical results of the behavioral evaluation: there were statistically significant in the differences between the MPI/AR group and the blank group(P<0.001)and in the differences between the MPI/AR group and in the captchilin group(p=0.024).There were not statistically significant in the differences between the MPI/AR group and acupoint catgut embedding group(P=0.375).But the behavioral evaluation of acupoint catgut embedding group was falling.There were statistically significant in the differences between acupoint catgut embedding group and the blank group(P=0.03).There were not statistically significant in the differences between acupoint catgut embedding group and the capsaicin group(P=0.145).But the behavioral evaluation of the capsaicin group was falling.There were not statistically significant in the differences between the capsaicin group and the blank group(P=0.083).But the behavioral evaluation of the blank group was falling.(3)The results of He stain:The AR group was the most inflammatory.The MPI/AR group was inferior.The acupoint embedded line group and the capsaicin group inflammatory situation were lighter than the MPI/AR group,and the white group was the lightest.(4)The results of Toluamine blue dyeing:The AR group were statisti--cally significant from the blank group(P=0.005)? acupoint catgut embedding group(P=0.003)and the capsaicin group(P=0.003).The MPI/AR group were statistically significant from the blank group(P=0.02)?acupoint catgut embedding group(P=0.016)and the capsaicin group(P=0.006).acupoint catgut embedding group were not statistically significant from the blank group(P=0.993)and the capsaicin group(P=1.00).the capsaicin group was not statistically significant from the blank group(P=0.878).(5)The results of immunization of SP:The AR group were statistically significant from The MPI/AR group(P=0.009).The MPI/AR group were statistically significant from the blank group(P<0.001)? acupoint catgut embedding group(P<0.001)and the capsaicin group(P=0.001).acupoint catgut embedding group were not statistically significant from the blank group(P=0.956)and the capsaicin group(P=0.912).the capsaicin group was not statistically significant from the blank group(P=0.956).(6)The results of Western blot test:The MPI/AR group were statistic--ally significant from the blank group(P=0.004)? acupoint catgut embedding group(P=0.008)and the capsaicin group(P=0.007).acupoint catgut embedding group were not statistically significant from the blank group(P=0.785)and the capsaicin group(P=0.975).the capsaicin group was not statistically significant from the blank group(P=0.809).(7)The results of Elisa test:The MPI/AR group were statistically significant from the blank group? acupoint catgut embedding group and the capsaicin group(P<0.001).acupoint catgut embedding group were not statistically significant from the blank group(P=0.547)and the capsaicin group(P=0.733).the capsaicin group was not statistically significant from the blank group(P=0.793).2.The effect of acupoint catgut embedding on the tolerance of OVA sensitizer.(1)the Statistical results of behavioral total score:The MPI/AR group had the highest behavioral score,followed by the sham embedding group and the acupoint embedding group.The rank-sum test showed that the differences of each group were statistically significant(P=0.012).By pairwise comparison of non-parametric test,the difference between MPI/AR group and acupoint catgut embedding group was statistically significant(P=0.009).There was no significant difference between the MPI/AR group and the sham embedding group(P=0.265).There was no significant difference between the acupoint embedding group and sham embedding group(P=0.634).(2)the Statistical results of concentration and time Tolerance:The mean tolerance concentration of acupoint catgut embedding group was higher than sham catgut embedding group and MPI/AR group.The average endurance time of acupoint wire embedding group was longer than that of sham wire embedding group and MPI/AR group.The statistical results of time tolerance showed that the differences between the groups were statistically significant(P=0.031).The difference between MPI/AR group B and acupoint catgut embedding group B was statistically significant(P=0.012).There was no significant difference between the MPI/AR group B and the sham catgut B group(P=0.497).There was no significant difference between acupoint catgut embedding group B and sham catgut embedding group B(P=0.051).Conclusion:1.Acupoint catgut implantation can improve the behavioral symptoms related to MPI/AR in SD rats.Alleviating the MPI/AR inflammation in the nasal mucosa of SD rats;Decreased the number of mast cells in nasal mucosa and SP content.2.Acupoint catgut implantation enhance the tolerance of SD rats to OVA in both concentration and time.
